目的 :研究海参糖胺聚糖 (hGAG)抗血栓机制。方法 :观察hGAG对内皮细胞的促凝活性、组织因子 (TF)、凝血酶调节蛋白 (TM )和纤溶酶原激活物抑制剂 1(PAI 1)的影响。结果 :细菌脂多糖(LPS)与内皮细胞孵育后 ,不同浓度的hGAG可使细胞促凝活性下降 ,TF抗原及mRNA转录下降 ,TM抗原表达及mRNA转录增强 ;hGAG可使内皮细胞培养上清液中的PAI 1抗原、活性、mRNA转录下降。结论 :hGAG通过下调内皮细胞TF表达 ,促进TM表达 ,降低内皮细胞PAI 1合成、分泌及抑制PAI 1mRNA转录 ,发挥抗血栓作用
Objective: To study the antithrombotic mechanism of glycosaminoglycan (hGAG) in sea cucumber. Methods: The effects of hGAG on the procoagulant activity of endothelial cells, tissue factor (TF), thrombin regulatory protein (TM) and plasminogen activator inhibitor 1 (PAI 1) were observed. Results: After cultured with lipopolysaccharide (LPS) and endothelial cells, different concentrations of hGAG decreased the procoagulant activity, decreased the expression of TF antigen and mRNA, increased the expression of TM antigen and mRNA transcripts; hGAG increased the activity of endothelial cell culture supernatant In the PAI 1 antigen, activity, mRNA transcription decreased. Conclusion: hGAG can reduce the expression of TF in endothelial cells, promote the expression of TM, decrease the PAI 1 synthesis, secretion and PAI 1 mRNA transcription in endothelial cells and exert antithrombotic effects
