目的观察痰瘀同治代表药物丹蒌片药物血清对血小板衍生因子BB型(PDGF-BB)诱导下体外培养的血管平滑肌(VSMC)增殖的影响,并探讨其可能的机制。方法 SD大鼠喂饲丹蒌片药物溶液收集药物血清。组织块贴壁法培养SD大鼠原代胸主动脉平滑肌细胞,取第3代细胞进行免疫荧光法鉴定,取4~7代细胞进行实验。MTT比色法观察正常血清培养及丹蒌片药物血清(低、中、高浓度)对PDGF-BB(25μg/L)所致的VSMC增殖的影响。结果 MTT法显示与对照组相比,药物血清组对PDGF-BB诱导的VSMC增殖呈剂量依赖性抑制,高浓度与低浓度比较有明显差异(P<0.05)。结论丹蒌片可有效抑制PDGF-BB诱导的VSMC增殖,提示其控制AS的过程可能与抑制平滑肌增殖并向内皮下迁移有关。
Objective To observe the effect of serum of Dan-ping tablet, a representative drug of phlegm and blood stasis, on the proliferation of vascular smooth muscle (VSMC) cultured in vitro induced by platelet-derived factor BB (PDGF-BB) and to explore its possible mechanism. Methods SD rats were fed with drug solution of danji tablet to collect drug serum. Tissue-block adherent method was used to culture primary thoracic aorta smooth muscle cells of SD rats. The third passage cells were identified by immunofluorescence method and the cells were cultured for 4-7 passages. MTT assay was used to observe the effect of normal serum culture and the drug serum (low, medium and high) on the proliferation of VSMC induced by PDGF-BB (25μg / L). Results MTT assay showed that compared with the control group, the drug serum group inhibited the proliferation of VSMC induced by PDGF-BB in a dose-dependent manner. There was significant difference between the high concentration and the low concentration (P <0.05). CONCLUSION Dan Dan tablet can effectively inhibit the proliferation of VSMC induced by PDGF-BB, suggesting that its process of controlling AS may be related to the inhibition of smooth muscle proliferation and its migration to the subcutaneous.
