目的:建立LC-ESI-MS法测定血浆中盐酸雷尼替丁的血药浓度。方法:采用Zorbax SB-C18(2.1×100 mm,3.5μm)色谱柱,流动相为乙腈-0.1%甲酸溶液(30:70)。电喷雾离子源(ESI),选择正离子模式检测,采用选择离子监测(SIM)方式进行定量分析,检测离子为m/z 315(雷尼替丁),m/z 180(内标非那西丁)。结果:血浆中盐酸雷尼替丁浓度在5~1000 ng/ml范围内线性关系良好,最低检测限为5 ng/ml。盐酸雷尼替丁的提取平均回收率为83.92%~103.5%,日内及日间RSD都低于12%。结论:该法操作简便、快速、灵敏度高,可用于雷尼替丁的临床血药浓度监测。
Objective: To establish a LC-ESI-MS method for determination of plasma concentration of ranitidine hydrochloride in plasma. Methods: Zorbax SB-C18 (2.1 × 100 mm, 3.5 μm) was used as the mobile phase. The mobile phase was acetonitrile-0.1% formic acid solution (30:70). Electrospray ionization (ESI), positive ion mode detection and quantitative ion-selective ionization (SIM) were used to detect ions with m / z 315 (ranitidine), m / z 180 Ding). Results: Linearity of ranitidine hydrochloride in plasma ranged from 5 to 1000 ng / ml. The lowest detection limit was 5 ng / ml. The average recoveries of ranitidine hydrochloride were 83.92% ~ 103.5%, with RSD less than 12% in both days and day. Conclusion: The method is simple, rapid and sensitive. It can be used to monitor the clinical plasma concentration of ranitidine.
