目的:探讨高浓度葡萄糖刺激下糖皮质激素对大动脉内皮细胞11β羟基类固醇脱氢酶(11β-HSD)基因表达的影响。方法:将猪髂动脉内皮细胞培养于高糖(30mmol/L)中,分别加入不同浓度糖皮质激素刺激,用RT-PCR法检测11β-HSD1,11β-HSD2 mRNA表达情况,Western-blot方法检测11β-HSD1,11β-HSD2蛋白表达情况。结果:正常对照组中,仅有少量的11β-HSD1 mRNA及蛋白表达,高糖状态下,11β-HSD1 mRNA和蛋白表达升高,且随着可的松浓度的增加而增加(P<0.05);高糖状态下,11β-HSD2 mRNA和蛋白表达无明显变化。结论:糖皮质激素使高糖培养下的动脉内皮细胞11β-HSD1表达增加,可能与糖尿病血管病变的发生发展有关。
OBJECTIVE: To investigate the effect of glucocorticoid on the gene expression of 11β-hydroxysteroid dehydrogenase (11β-HSD) in aorta endothelial cells under high glucose. Methods: The porcine iliac artery endothelial cells were cultured in high glucose (30mmol / L) and stimulated with different concentrations of glucocorticoid respectively. The expression of 11β-HSD1, 11β-HSD2 mRNA was detected by RT-PCR and Western blot 11β-HSD1,11β-HSD2 protein expression. Results: In normal control group, only a small amount of 11β-HSD1 mRNA and protein were expressed. Under high glucose condition, the mRNA and protein expression of 11β-HSD1 increased, and increased with the increase of cortisone concentration (P <0.05) There was no significant change of 11β-HSD2 mRNA and protein expression in high glucose. Conclusion: Glucocorticoid can increase the expression of 11β-HSD1 in arterial endothelial cells under high glucose, which may be related to the occurrence and development of diabetic vascular disease.
