潜阳育阴颗粒对自发性高血压大鼠肾脏氧化应激及炎症因子的影响

目的:研究潜阳育阴颗粒对自发性高血压大鼠肾脏氧化应激及炎症因子的影响,并探讨其改善自发性高血压的作用机制。方法:自发性高血压大鼠,随机分为模型组、潜阳育阴颗粒组、贝那普利组及潜阳育阴颗粒+贝那普利组,分别给予潜阳育阴颗粒混悬液(5 g·kg-1),贝那普利混悬液(1.67 mg·kg-1),潜阳育阴颗粒混悬液(5 g·kg-1)+贝那普利混悬液(1.67 mg·kg-1)ig,另设正常血压(WKY)大鼠为正常组,连续给等容量药8周后处死大鼠收集肾脏组织。观察大鼠血压、微量白蛋白和肌酐的变化,蛋白质免疫印迹(Western blot)检测肾脏局部NADPH氧化酶-4(NOX4)的水平,核转录因子-κB(NF-κB)p65及肿瘤坏死因子-α(TNF-α),白细胞介素-6(IL-6)蛋白表达量的变化,并观察潜阳育阴颗粒对这些指标释放量的影响。结果:与正常组比较,模型组大鼠血压、微量白蛋白和肌肝明显升高,肾脏组织NOX4,NF-κB,TNF-α和IL-6蛋白表达明显升高(P<0.01);潜阳育阴颗粒能抑制模型组肾脏NOX4的表达,降低NF-κB p65,TNF-α,IL-6蛋白表达量,降低血压、微量白蛋白和肌肝的含量(P<0.01),而潜阳育阴颗粒和贝纳普利两药协同的效果更加明显(P<0.05)。结论:潜阳育阴颗粒改善自发性高血压大鼠肾损害机制可能与抑制氧化应激进而减少炎症因子释放相关。 Objective: To study the effect of QianYangYuYin Granules on renal oxidative stress and inflammatory factors in spontaneously hypertensive rats and to explore its mechanism of action to improve spontaneous hypertension. Methods: Spontaneous hypertensive rats were randomly divided into 4 groups: model group, Qiangyuyuanyin granule group, benazepril group and Qiangyouyuanyin granule + benazepril group, respectively. (5 g · kg -1), benazepril suspension (1.67 mg · kg -1), Qiangyuxuanyin granule suspension (5 g · kg -1) + benazepril suspension 1.67 mg · kg-1) ig, another normal blood pressure (WKY) rats were normal group, and rats were sacrificed 8 weeks after the equal volume of drug was collected to collect the kidney tissue. The changes of blood pressure, microalbumin and creatinine were observed. The levels of NADPH oxidase - 4 (NOX4), nuclear factor kappa B (NF - κB) p65 and tumor necrosis factor - α (TNF-α), interleukin-6 (IL-6) protein expression changes, and observe the effects of QianYangYuYin particles on the release of these indicators. Results: Compared with the normal group, the levels of blood pressure, microalbumin and muscular liver in model group were significantly increased, and the expressions of NOX4, NF-κB, TNF-α and IL-6 in renal tissue were significantly increased (P <0.01) Yangyin Granule could inhibit the expression of NOX4, decrease the expression of NF-κB p65, TNF-α and IL-6, decrease the content of blood pressure, microalbumin and muscle liver in model group (P <0.01) The combination of Yinyin granule and benazepril synergistic effect is more obvious (P <0.05). Conclusion: The mechanism of Qiangyou-Yin-Yin Granule in improving renal damage in spontaneously hypertensive rats may be related to the inhibition of oxidative stress and the reduction of the release of inflammatory cytokines.
