目的:研究胃癌中环氧合酶2(COX-2)的表达情况和生物学意义,以及与核因子κB(NF-κB)的相关性。方法:选择胃癌及癌旁正常组织41例,对COX-2、NF-κB进行RT-PCR及Western blot检测。结果:41例胃癌标本中有28例COX-2阳性,26例NF-κB阳性,COX-2和NF-κB表达呈正相关。COX-2和NF-κB基因表达与年龄、性别、大小、组织学类型无关,但与浸润深度、临床分期和肿瘤的淋巴结转移具有显著的相关性(P<0.05)。结论:COX-2过表达在肿瘤侵袭过程中发挥着重要作用,而NF-κB可能通过调节COX-2的表达在胃癌细胞增殖过程中起重要作用。
Objective: To investigate the expression and biological significance of cyclooxygenase 2 (COX-2) in gastric cancer and its relationship with nuclear factor κB (NF-κB). Methods: Twenty-one cases of gastric cancer and adjacent normal tissues were selected for RT-PCR and Western blot analysis of COX-2 and NF-κB. Results: Of 41 gastric cancer samples, 28 were COX-2 positive, 26 were NF-κB positive, and COX-2 and NF-κB were positively correlated. The expression of COX-2 and NF-κB was not related to age, sex, size and histological type, but significantly correlated with the depth of invasion, clinical stage and lymph node metastasis (P <0.05). CONCLUSION: Overexpression of COX-2 plays an important role in tumor invasion. NF-κB may play an important role in the proliferation of gastric cancer cells by regulating the expression of COX-2.
