在前期巴西橡胶树胶乳转录组测序的基础上,利用qRT-PCR技术,从巴西橡胶树中克隆得到1个NAC转录因子基因,命名为HbNAC55.HbNAC55全长1 755 bp,该基因编码584个氨基酸,蛋白质分子量为65.06ku,等电点为4.79.蛋白结构分析发现,该蛋白含有一个NAM结构域,具有典型的NAC类蛋白的结构特征,与木薯、胡杨、可可、土瓶草、梅、胡桃、甜橙等物种的NAC家族成员具有较高的同源性,分别为81%、61%、56%、57%、55%和53%.通过qRT-PCR发现HbNAC55在橡胶树胶乳中表达量最高.茉莉酸和乙烯均能诱导胶乳中HbNAC55表达上调,其中茉莉酸甲酯处理12h后HbNAC55表达量提高了近6倍,乙烯处理48 h后HNAC55表达量提高了近1.7倍.本研究将为进一步研究HbNAC55在巴西橡胶树中的功能提供了帮助.“,”Based on the latex transcriptome sequencing of the Hevea brasiliensis,a NAC transcription factor gene named HbNA C55 was cloned from Hevea brasiliensis by qRT-PCR technology.HbNA C55 was 1 755 bp in full length,encoding 584 amino acids with molecular mass of 65.06 ku and pI of 4.79.The protein structure analysis revealed that HbNA C55 contained a NAM domain with typical structural features of class NAC proteins.which showed high identity of 81%,61%,56%,57%,55% and 53% to those of the NAC proteins from Manihot esculenta,Populus euphratica,Theobrom cacao,Prunus mumes,Juqhans reqia and Citrus sinensis,qRT-PCR found that the expression of HbNA C55 was the highest in the latex of Hevea brasiliensis.Both jasmonic acid and ethylene could induce up the regulation of HbNA C55 expression in the latex,and the expression of HbNA C55 was increased nearly 6 times and 1.7 times after methyl jasmonate treatment for 12 h and ethylene treatment for 48 h,respectively,These results might provid help for further study on the function of HbNA C55 in Hevea brasiliensis.
