目的:探讨西洋参茎叶二醇三醇组皂甙(PQTS)对大鼠离体工作心脏血流动力学的影响及其作用机理。方法:制备大鼠离休工作心脏,观察三种剂量(30,100和200mg/L)PQTS的作用。提高细胞外液Ca2+浓度(0.56g/L)观察Ca2+对100mg/LPQTS作用的影响。另外向灌流液中加入异丙肾上腺素(Isop1×10-9g/L),观察IsoP对100mg/l。PQTS作用的影响。结果:PQTS具有负性肌力和负性频率作用,并呈现明显的量效关系。100mg/LPQTS可以拮抗因Ca2+浓度增加所致的正性肌力作用,但不能对抗Isop所致的正性肌力作用。结论:PQTS具有负性肌力和负性频率作用。其机理可能是因抑制Ca2+内流所致,而与β-受体无关。
Objective: To investigate the effect and mechanism of PQTS on hemodynamics of isolated working heart in rats. Methods: The working heart of rats was retired, and the effects of three doses of PQTS (30, 100 and 200 mg / L) were observed. Conclusion: PQTS has negative inotropic and negative frequency effects. The mechanism may be due to inhibition of Ca2 + influx, but not with the β-receptor.
