目的 观察白细胞介素-1(IL-1)和L-单甲基精氨酸(L-NMMA)对兔髁状突软骨细胞一氧化氮(NO)合成和诱导型一氧化氮合酶(iNOS)基因表达的影响。方法 用硝酸还原酶法和RT-PCR反应分别检测软骨细胞培养上清液中NO浓度和细胞内iNOS mRNA表达的变化。结果 单独使用IL-1,软骨细胞培养液中NO水平和iNOS表达水平显著升高,配伍加入L-NMMA能有效降低由于IL-1刺激引起的NO水平升高,但不能降低iNOS表达的升高幅度。结论 IL-1能显著增加体外培养的兔髁状突软骨细胞iNOS的表达而促进NO的生成,而L-NMMA对这种效应有明显抑制作用。
Objective To observe the effects of interleukin-1 (IL-1) and L-NMMA on the synthesis of nitric oxide (NO) and inducible nitric oxide synthase (iNOS) in rabbit condylar chondrocytes ) Gene expression. Methods Nitric acid reductase and RT-PCR were used to detect the changes of NO concentration and intracellular iNOS mRNA expression in chondrocyte culture supernatants. Results IL-1 alone increased the expression of NO and iNOS in chondrocytes, and the addition of L-NMMA could effectively decrease the level of NO induced by IL-1 stimulation, but not decrease the expression of iNOS Magnitude. Conclusion IL-1 can significantly increase the iNOS expression in rabbit condylar chondrocytes and promote the production of NO, but L-NMMA can significantly inhibit this effect.
