目的探讨人类重组粒—巨噬细胞集落刺激因子(rhGM-CSF)对辐射物免疫器官的AgNOR定量论功.方法雄性Beagle狗14只,随机分成辐射时抵组4只.高、低剂量rhGM-CSF治疗组各5只.动物均双侧髂骨屏蔽,其它部位用60CO照射.除对照组外.其它两组股静脉注射rhGM-CSF共20d,低剂量组为10μg·kg-1·d-1,高剂量组为20μg·kg-1·d-1,第30d,取出脾脏与淋巴结,组织切片,Ploton’s法AgNOR染色、用分析仪计数100个细胞核内AgNon颗粒数,求其均值。结果低剂量组rhGM-CSF组的AgNOR颗粒数高于对照组(P<0.05),高剂量组rhGM-CSF的AgNOR颗粒数明显高于对照组(P<0.01).结论rhGM—CSF可促进辐射后狗脾及淋巴结形态与功能的恢复,其恢复程度与该因子剂量有关.
Objective To investigate the quantitative determination of AgNOR in irradiated immune organs by recombinant human granulocyte-macrophage colony-stimulating factor (rhGM-CSF). Methods Fourteen male Beagle dogs were randomly divided into four groups. High and low doses of rhGM-CSF treatment group of 5. Animals were bilateral iliac shield, 60CO irradiation of other parts. In addition to the control group. The other two groups were given rhGM-CSF for 20 days, the low dose group was 10μg · kg-1 · d-1 and the high dose group was 20μg · kg-1 · d-1. Results The number of AgNOR particles in the rhGM-CSF group was significantly higher than that in the control group (P <0.05). The number of AgNOR particles in the rhGM-CSF group was significantly higher than that of the control group (P <0.01). Conclusion rhGM-CSF can promote the recovery of the morphology and function of dog, spleen and lymph node after radiation, and the degree of recovery is related to the dosage of this factor.
