目的:以孕前应激诱导建立产后抑郁(PPD)小鼠模型,观察越鞠丸醇提物(YJ)对该模型的快速抗抑郁作用。方法:将40只BALB/c雌鼠随机分为2组,空白对照组不予任何刺激,孕前应激组给予慢性束缚刺激。3周后孕前应激组小鼠雌雄合笼交配怀孕,于分娩后3周检测模型组母鼠行为学,包括蔗糖偏好测试(SPT)、强迫游泳测试(FST)、悬尾测试(TST)及陌生环境摄食实验(NSFT)。造模成功后,将模型组随机分为4组:模型组、越鞠丸醇提物组、氯胺酮组、氟西汀组。越鞠丸醇提物及氯胺酮为单次给药,氟西汀为连续21d给药。结果:产后3周,与空白对照组相比,孕前应激诱导的产后抑郁组模型母鼠在糖水偏好测试中的蔗糖消耗百分比显著降低(P<0.001),在强迫游泳测试和悬尾测试中绝望状态不动时间显著增加(P<0.01,P<0.01),在陌生环境摄食实验中摄食潜伏时间明显增加而单位时间摄食量显著减少(P<0.01,P<0.01)。单次给予YJ和氯胺酮24h之后,模型组母鼠的糖水消耗百分比显著增加(P<0.01),强迫游泳和悬尾测试的绝望不动时间明显降低(P<0.01,P<0.01),陌生环境摄食测试中潜伏时间显著缩短且单位时间摄食量明显增加(P<0.01,P<0.01),而氟西汀组未显示相同作用。结论:氟西汀连续给药对于孕前应激诱导的PPD模型小鼠无明显治疗作用,提示该模型可能属于难治性抑郁模型;在此模型上,越鞠丸具有与西药氯胺酮一致的快速抗抑郁作用。
OBJECTIVE: To establish postpartum depression (PPD) mouse model by pre-pregnancy stress and to observe the rapid anti-depressant effect of Yueju pill alcohol extract (YJ) on this model. Methods: Forty female BALB / c mice were randomly divided into two groups. The blank control group received no stimulation and the pre-pregnancy stress group received chronic restraint stimulation. Three weeks after pregnancy, the mice in the pre-pregnancy stress group were mated with the male and female cages, and the behavioral models including the sucrose preference test (SPT), forced swimming test (FST), tail-hanging test (TST) Strange Environment Ingestion Experiment (NSFT). After successful modeling, the model group was randomly divided into 4 groups: model group, Yueju pills alcohol extract group, ketamine group, fluoxetine group. Yueju pills alcohol extract and ketamine for a single administration, fluoxetine for 21d administration. Results: Compared with the blank control group, the percentage of sucrose consumption in the postpartum stress-induced postpartum depression model rats was significantly lower (P <0.001) in the sugar preference test at 3 weeks postpartum. In the forced swimming test and the tail suspension test (P <0.01, P <0.01). In the unfamiliar environment, the latency to ingest food significantly increased and the food consumption per unit time significantly decreased (P <0.01, P <0.01). After a single administration of YJ and ketamine for 24 h, the percentage of sugar consumption of model rats increased significantly (P <0.01), forced swimming and suspension test decreased significantly (P <0.01, P <0.01), unfamiliar environment Latent time during feeding test was significantly shortened and food consumption per unit time significantly increased (P <0.01, P <0.01), while the fluoxetine group did not show the same effect. CONCLUSION: Continuous administration of fluoxetine has no obvious therapeutic effect on pre-pregnancy stress-induced PPD mice, suggesting that this model may belong to the model of refractory depression. In this model, Yueju pills have the fast anti-ketamine Depression.
