目的:比较自然衰老法与卵巢去势法建立的ICR小鼠围绝经期模型,并对模型进行评价。方法:通过观察阴道脱落细胞,比较自然衰老组及卵巢去势组小鼠的动情周期情况;观察小鼠悬尾活动,比较自然衰老组及卵巢去势组小鼠的情绪状态;测定血清性激素,比较血清卵泡刺激素(FSH)、促黄体激素(LH)、雌二醇(E2)的含量;测定小鼠下丘脑神经肽,比较下丘脑Kiss-1及其受体GPR54 mRNA的表达。结果:自然衰老组及卵巢去势组小鼠动情周期均紊乱,动情频率均减少;两组小鼠悬尾活动比均显著提高;两组小鼠血清的FSH、LH水平显著上升,E2水平明显降低;两组小鼠的下丘脑Kiss-1及受体GPR54 mRNA的表达显著升高。相比卵巢去势小鼠,自然衰老小鼠动情周期相对延长,动情频率明显减少,血清的FSH、LH水平上升更显著,E2水平降低更显著,Ki SS-1/GPR54 mRNA的表达升高更明显。结论:自然衰老法与卵巢去势法均能建立小鼠围绝经期模型,而自然衰老法优于卵巢去势法。
OBJECTIVE: To compare the perimenopausal model of ICR mice established by natural aging and ovariectomized methods and to evaluate the model. Methods: By observing vaginal cells, the estrous cycle of natural aging group and ovariectomized group were compared. The mice tail suspension activity was observed to compare the emotional state of natural aging group and ovariectomized group. The levels of serum sex hormones, The contents of FSH, LH and E2 were compared. The hypothalamus neuropeptide was measured and the expression of KissR1 and its receptor GPR54 mRNA in the hypothalamus was compared. Results: The estrous cycle was disordered and the frequency of estrus was decreased in both the natural aging group and the ovariectomized group. The tail suspension activities of the two groups were significantly increased. The levels of serum FSH and LH were significantly increased and the level of E2 was significantly increased Decreased; expression of Kiss-1 and receptor GPR54 mRNA in hypothalamus of both groups increased significantly. Compared with ovariectomized mice, the estrous cycle of naturally aged mice was relatively prolonged, the frequency of estrus was significantly reduced, serum FSH, LH levels increased more significantly, E2 levels decreased more significantly, Ki SS-1 / GPR54 mRNA expression increased more obvious. Conclusion: Both natural aging and ovariectomized mice can establish mouse perimenopausal model, while natural aging is superior to ovariectomized.
