目的研究XRCC1、XPD基因多态性及交互作用与铅毒性易感性的关系。方法采集326名某蓄电池企业铅作业工人静脉血和晨尿,检测血铅、血锌原卟啉(ZPP)和尿铅,用PCR-RFLP检测XRCC1、XPD的基因多态性,分析不同基因型及交互作用对铅毒性易感性的影响。结果 XRCC1-194CT+TT基因组平均血铅值较XRCC1-194CC组高;病例组XRCC1-194CT+TT基因型比例大于对照组,慢性铅中毒发生率风险较XRCC1-194CC组高。结论在相同职业铅暴露环境下,XRCC1-194CT+TT基因型工人对铅毒性易感;XPD Asp312Asn和Lys751Gln多态性与铅毒性易感性无关;XRCC1Arg194Trp、XPD Asp312Asn和Lys751Gln基因对铅毒性的影响不存在交互作用。
Objective To study the relationship between polymorphisms of XRCC1 and XPD genes and their interaction with susceptibility to lead toxicity. Methods Venous and morning urine were collected from 326 lead workers in a storage battery. Blood lead, zinc protoporphyrin (ZPP) and urinary lead were detected. The polymorphisms of XRCC1 and XPD were detected by PCR-RFLP. The genotypes of different genotypes And the effect of interaction on susceptibility to lead toxicity. Results The average blood lead level of XRCC1-194CT + TT group was higher than that of XRCC1-194CC group. The proportion of XRCC1-194CT + TT genotype was higher in case group than in control group, and the risk of chronic lead poisoning was higher than that of XRCC1-194CC group. Conclusion XRCC1-194CT + TT genotype workers are susceptible to lead toxicity in the same occupational exposure to lead. The polymorphisms of XPD Asp312Asn and Lys751Gln have no relationship with susceptibility to lead toxicity. The effects of XRCC1Arg194Trp, XPD Asp312Asn and Lys751Gln genes on lead toxicity are not related to lead toxicity There is interaction.
