目的:建立哈萨克药鹿草根中β-蜕皮甾酮的含量测定方法。方法:采用反相,使用Waters XTerra RP C18(250 mm×4.6 mm,5μm)色谱柱,以甲醇-1%醋酸溶液(27∶73)为流动相,体积流量为1.0 mL.min-1,检测波长为250 nm,柱温30℃。结果:在上述条件下,β-蜕皮甾酮浓度在1.45~14.5μg.mL-1范围内呈良好的线性关系(r=0.9998);平均加样回收率(n=9)为100.4%。结论:本方法准确,灵敏度高,重现性好,能有效测定鹿草根中β-蜕皮甾酮含量。
Objective: To establish a method to determine the content of β-ecdysterone in the root of Kazak herb. Methods: The mobile phase was reverse-phase HPLC with a mobile phase of methanol-1% acetic acid (27:73) at a flow rate of 1.0 mL · min-1 on a Waters XTerra RP C18 (250 mm × 4.6 mm, Wavelength of 250 nm, column temperature 30 ℃. Results: Under the above conditions, the β-ecdysterone concentration showed a good linear relationship (r = 0.9998) in the range of 1.45-14.5μg.mL-1; the average recovery was 100.4%. Conclusion: The method is accurate, sensitive and reproducible. It can effectively determine the β-ecdysterone content in Deer grass roots.
