目的:观察筋骨片对小鼠的急性毒性和对大鼠的长期毒性,为临床用药的安全剂量提供参考。方法:①急性毒性试验:昆明种小鼠40只,随机分成给药组和对照组,采用最大给药量法测定小鼠口服筋骨片最大耐受量;②长期毒性试验:SD大鼠120只,随机分成4组,采用3个剂量(按生药量计为61.4,30.7,15.3 g/kg)ig大鼠。每天给药1次,连续ig6个月,观察大鼠的一般情况,检测血常规及血液生化学、组织病理变化及停药1个月时上述指标的变化。结果:小鼠口服筋骨片的最大耐受量按生药计为842 g/kg,相当于成人临床每日口服剂量的1 477倍。大鼠连续给药6个月后筋骨片各剂量组各项指标未见明显异常改变;2只大鼠的肝、肾细胞有轻度水样变性,1只大鼠的细小支气管周围淋巴细胞浸润。停药1个月后,大鼠体重、摄食量、行为活动、血象、重要器官脏器系数均正常,病理学检查,各组各脏器均未发现明显的改变。结论:筋骨片在规定剂量下服用是安全可靠的。

Six months after continuous administration, there were no obvious abnormal changes in each dose of each group. The liver and kidney cells of two rats were mildly watery degeneration, and one rat had small bronchial peripheral lymphocyte infiltration . One month after discontinuation of treatment, the body weight, food intake, behavioral activity, hemogram and the vital organ coefficients of rats were all normal. No significant changes were found in each organ of each group. Conclusion: It is safe and reliable to use bones and bones tablets under the prescribed dosage.
