目的探讨熊果酸(UA)对ER(-)、Her-2过表达的人乳腺癌SK-BR-3细胞增殖和凋亡的影响。方法利用噻唑蓝(MTT)比色法观察UA对SK-BR-3细胞增殖的影响。用Hoechst 33258荧光染色及电子显微镜观察凋亡细胞核形态变化。流式细胞仪检测UA对细胞周期的影响;DNA琼脂糖凝胶电泳观察DNA片段的断裂。结果UA明显抑制SK-BR-3细胞的增殖,差异有统计学意义(P<0.05),并呈时间和浓度依赖性。经UA处理后的实验组细胞呈现典型的凋亡核固缩表现,胞核呈致密浓染的颗粒状或块状荧光。流式细胞仪检测发现UA作用48h,后SK-BR-3细胞阻滞在G0/G1期,差异有统计学意义(均P<0.05)。DNA琼脂糖凝胶电泳显示DNA大片段的断裂。结论UA在体外对SK-BR-3细胞具有抗增殖和诱导凋亡的作用。
Objective To investigate the effects of ursolic acid (UA) on proliferation and apoptosis of human breast cancer SK-BR-3 cells overexpressing ER (-) and Her-2. Methods The effect of UA on the proliferation of SK-BR-3 cells was observed by MTT assay. Morphological changes of apoptotic nuclei were observed with Hoechst 33258 fluorescence staining and electron microscopy. Flow cytometry was used to detect the effect of UA on the cell cycle. DNA fragmentation was observed by DNA agarose gel electrophoresis. Results UA significantly inhibited the proliferation of SK-BR-3 cells, the difference was statistically significant (P <0.05), and in a time and concentration-dependent manner. After UA treatment, the cells in the experimental group showed typical apoptotic nucleus pyknosis, and the nuclei showed dense and dense granular or massive fluorescence. The results of flow cytometry showed that after UA treatment for 48h, SK-BR-3 cells arrested in G0 / G1 phase, the difference was statistically significant (P <0.05). DNA agarose gel electrophoresis shows fragmentation of large DNA fragments. Conclusion UA has anti-proliferation and apoptosis-inducing effects on SK-BR-3 cells in vitro.
