探讨香菇多糖对于脓毒血症的预防与恢复,分析香菇多糖的保护作用并且从肿瘤坏死因子(TNF-α)水平上探索其保护机制。将40只健康Wistar雄性大鼠随机分成4组:A组(假手术组)、B组(脓毒血症组)、C组(香菇多糖预防组)、D组(香菇多糖治疗组)。手术之前A组、B组以及D组给予尾静脉注射生理盐水0.2 mL/kg,C组尾静脉注射香菇多糖注射液0.1 g/kg,均1周注射2次,持续4周。4周后,A组进行假手术;B组、C组以及D组进行手术。利用盲肠结扎穿刺法制备脓毒血症大鼠模型成功后,给予D组进行尾静脉注射香菇多糖注射液0.1 mg/kg,A组、B组和C组进行生理盐水注射0.2 mL/kg,12 h后采用ELISA法检测各组大鼠血清中TNF-α的含量。香菇多糖干预后,C组和D组内毒素水平明显低于B组(P=0.047,0.006);B组、C组和D组的内毒素水平明显高于A组(P=0.023,0.028,0,035);A组、C组、D组血清TNF-α水平低于B组(P=0.001,0.022,0.018);C组和D组血清TNF-α水平高于A组(P=0.009,0.012)。香菇多糖对脓毒血症有良好的预防和恢复作用,且能明显降低脓毒血症大鼠血清中TNF-α的含量,这可能是其保护脓毒血症的重要作用机制之一。
To explore the prevention and recovery of lentinan by lentinans, analyze the protective effect of lentinan and explore its protective mechanism from the level of tumor necrosis factor (TNF-α). Forty healthy Wistar male rats were randomly divided into 4 groups: group A (sham operation group), group B (sepsis group), group C (lentinan prevention group) and group D (lentinan treatment group). Group A, group B and group D received 0.2 mL / kg of saline injection into tail vein and group C received 0.1 g / kg of lentinan injected intravenously twice a week for 4 weeks. After 4 weeks, group A underwent sham operation; group B, group C and group D underwent surgery. After the successful sepsis rat model was established by cecal ligation and puncture, group D was given intravenous injection of lentinan 0.1 mg / kg, Group A, B and C were given saline 0.2 mL / kg, h after the ELISA method was used to detect the content of serum TNF-α in each group. Endotoxin levels in groups C and D were significantly lower than those in group B after intervention of lentinan (P = 0.047,0.006); endotoxin levels in groups B, C and D were significantly higher than those in group A (P = 0.023,0.028, 0,035). The levels of serum TNF-α in group A, group C and group D were lower than those in group B (P = 0.001,0.022,0.018), while those in group C and D were significantly higher than those in group A (P = 0.009,0.012 ). Lentinan can prevent and recover sepsis effectively, and can significantly reduce the level of TNF-α in serum of sepsis rats, which may be one of its important mechanisms of protecting sepsis.
