目的:通过检测肝癌细胞系HepG2和HepG2.2.15细胞中基质金属蛋白酶-2和-9的表达水平及两种细胞的生长速度,探讨MMPs在HBV的影响下对肝细胞癌增殖能力的影响及其与肿瘤侵袭转移之间的关系。方法:用免疫组化法检测HepG2细胞及转染了HBV表达质粒的HepG2.2.15细胞中MMP-2和MMP-9的表达情况,用四甲基偶氮唑盐比色法(MTT法)检测细胞增殖情况并观察两种细胞的某些生物学活性。结果:MTT及细胞生长曲线显示HepG2细胞的生长速度快于HepG2.2.15细胞。免疫组化检测HepG2细胞中MMP-9的表达高于HepG2.2.15细胞,而MMP-2的表达在两种细胞中均不明显,无显著性差异。结论:通过体外检测肝细胞癌细胞系HepG2细胞和HepG2.2.15细胞生长状态的差异,揭示HBV对肝细胞癌的生长、增殖具有一定的抑制作用,HBV能够抑制肝细胞癌中MMP-9的表达,从而间接影响了其侵袭和转移的能力。
OBJECTIVE: To investigate the effect of MMPs on the proliferation of hepatocellular carcinoma (HCC) under the influence of HBV by detecting the expression of MMP-2 and -9 and the growth rate of both cells in HepG2 and HepG2.2.15 cells And tumor invasion and metastasis of the relationship between. Methods: The expressions of MMP-2 and MMP-9 in HepG2 cells and HepG2.2.15 cells transfected with HBV plasmids were detected by immunohistochemistry and detected by MTT assay Cell proliferation was observed and some of the biological activities of both cells were observed. Results: MTT and cell growth curve showed HepG2 cells grew faster than HepG2.2.15 cells. Immunohistochemistry showed that the expression of MMP-9 in HepG2 cells was higher than that in HepG2.2.15 cells, while the expression of MMP-2 was not obvious in both cells, and there was no significant difference. Conclusion: The differences of the growth of HepG2 cells and HepG2.2.15 cells in vitro were detected by HE staining, which showed that HBV could inhibit the growth and proliferation of Hepatocellular carcinoma. HBV could inhibit the expression of MMP-9 in hepatocellular carcinoma , Which indirectly affect their ability to attack and transfer.
