目的 从猪骨胶原蛋白中提取小肽做为血管紧张素转换酶抑制剂。方法 猪骨胶原蛋白经胰蛋白酶水解 ,阳离子交换树脂层析 ,Sephadex G2 5凝胶过滤 ,反相高效液相层析 (HPLC) ,最终得到一种 9肽 ,其氨基酸组成 :Ile,Ser,His,Gly ,Ala,Pro ,Tyr,Leu ,Asp。结果 以马尿酰 甘氨酰 甘氨酸 (HGG)为底物 ,对从猪肺中提取的血管紧张素转换酶 (ACE)有明显的抑制作用 ,I50为 2 .6× 1 0 - 2 mmol/L。将含此肽 0 .0 1 %的混合物做静脉注射试验 ,对肾性高血压大鼠和自发性高血压大白鼠均有明显降压作用 ;口服给药 1 5d ,对自发性高血压有一定的降压作用。对离体豚鼠回肠平滑肌有收缩作用。结论 胶原蛋白酶解物 9肽的抗高血压效果 ,可能是通过对血管紧张素转换酶的抑制作用。
Objective To extract small peptides from porcine collagen as angiotensin converting enzyme inhibitors. Methods The porcine collagen was purified by trypsin hydrolysis, cation exchange resin chromatography, Sephadex G2 5 gel filtration and reversed-phase high performance liquid chromatography (HPLC) to obtain a 9 peptide with the amino acid composition of Ile, Ser, His , Gly, Ala, Pro, Tyr, Leu, Asp. Results HGG was used as a substrate to inhibit angiotensin converting enzyme (ACE) extracted from porcine lung. The I50 was 2.6 × 10 -2 mmol / L . Will contain 0.1% of this peptide mixture to do intravenous injection test, renal hypertensive rats and spontaneously hypertensive rats were significantly antihypertensive effect; oral administration of 15d, there is a certain spontaneous hypertension Antihypertensive effect. On isolated guinea pig ileum smooth muscle contraction. Conclusion The antihypertensive effect of collagenase 9 peptide may be through inhibition of angiotensin converting enzyme.
