目的探讨不同浓度葡萄糖对人牙龈成纤维细胞分泌Ⅰ(COL1)、Ⅲ型胶原蛋白(COL3)及纤维粘连蛋白(FN)的影响。方法分别于5.5、7.0、8.8、10.0、15.0、20.0 mmol/L的葡萄糖溶液中培养人牙龈成纤维细胞3~7 d,酶联免疫吸附试验(ELISA)法测定其分泌COL1、COL3及FN的量。结果人牙龈成纤维细胞培养3 d后,葡萄糖浓度高于10.0 mmol/L时,COL1的分泌量减少;高于15.0 mmol/L时,COL3的分泌量减少;FN与COL3情况相似。培养7 d后,葡萄糖浓度高于10.0 mmol/L时,COL1、COL3及FN的分泌量均减少。结论随着葡萄糖浓度的增高,人牙龈成纤维细胞分泌COL1、COL3及FN的量均有减少趋势,且高糖条件下更为明显。
Objective To investigate the effects of different concentrations of glucose on the secretion of human gingival fibroblasts COL1, COL3 and fibronectin (FN). Methods Human gingival fibroblasts were cultured in glucose solution of 5.5, 7.0, 8.8, 10.0, 15.0 and 20.0 mmol / L for 3-7 d, respectively. The secretion of COL1, COL3 and FN was measured by enzyme linked immunosorbent assay (ELISA) the amount. Results After 3 days of human gingival fibroblasts cultured, the secretion of COL1 decreased when glucose concentration was higher than 10.0 mmol / L. When the concentration was higher than 15.0 mmol / L, the secretion of COL3 decreased; FN was similar to COL3. After 7 days of culture, the secretion of COL1, COL3 and FN decreased when glucose concentration was higher than 10.0 mmol / L. Conclusions With the increase of glucose concentration, the amount of COL1, COL3 and FN secreted by human gingival fibroblasts tended to decrease, especially under high glucose conditions.
