目的观察大鼠戊四氮癫痫模型海马齿状回神经元凋亡情况及凋亡诱导因子(apoptosis-inducing factor,AIF)、胱天蛋白酶(Caspase)-3阳性表达的意义。方法将12只Wistar大鼠随机分为模型组和空白对照组,每组6只。模型组大鼠腹腔注射戊四氮,空白对照组大鼠腹腔注射生理盐水。造模成功后观察模型组与空白对照组大鼠海马齿状回组织切片尼氏染色及AIF、Caspase-3蛋白表达的情况。结果空白对照组和模型组大鼠海马齿状回区尼氏染色400倍镜下单个视野神经细胞计数分别为(99.76±11.89)、(78.69±10.94)个,Caspase-3灰度值分别为154.81±16.06、131.65±16.81,AIF蛋白灰度值分别为173.09±9.57、158.34±6.33,组间比较差异均有统计学意义(P<0.05)。结论依赖Caspase的Caspase-3和不依赖Caspase的AIF两种凋亡途径均参与了癫痫导致的神经细胞凋亡过程。
Objective To observe the apoptosis and the expression of apoptosis-inducing factor (AIF) and caspase-3 in rat hippocampal dentate gyrus neurons after pentylenetetrazole-induced epilepsy. Methods Twelve Wistar rats were randomly divided into model group and blank control group, with 6 rats in each group. Rats in model group were injected intraperitoneally with pentylenetetrazol, while those in blank control group were injected intraperitoneally with normal saline. After successful modeling, Nissl staining and expression of AIF and Caspase-3 protein in hippocampal dentate gyrus of model group and blank control group were observed. Results The number of single visual field neurons in the control group and the untreated group were (99.76 ± 11.89) and (78.69 ± 10.94), respectively. The gray values of Caspase-3 were 154.81 ± 16.06, 131.65 ± 16.81, respectively. The gray value of AIF protein was 173.09 ± 9.57 and 158.34 ± 6.33, respectively. There was significant difference between the two groups (P <0.05). Conclusion Both Caspase-dependent Caspase-3 and Caspase-independent AIF pathways are involved in the process of neuronal apoptosis induced by epilepsy.
