目的研究肿瘤抑素(Tumstatin)185-203位氨基酸(19肽)对人卵巢癌细胞(SKOV3)的抑制作用,探讨其抑瘤作用机制。方法使用免疫组化、TUNEL方法检测肿瘤抑素19肽对SKOV3细胞增殖和凋亡的影响,以及caspase 3蛋白的表达情况。结果 19肽组抑瘤率可达36.47%,肿瘤细胞有明显的凋亡,caspase3蛋白的表达显著增加,与对照组比较差异有统计学意义(P<0.05)。结论肿瘤抑素19肽可以明显地抑制裸鼠卵巢癌细胞的生长,其抑制卵巢癌的作用机制可能与其激活caspase通路有关。
Objective To study the inhibitory effect of Tumstatin amino acid 185-203 (19 peptide) on human ovarian cancer cells (SKOV3) and to investigate its anti-tumor mechanism. Methods Immunohistochemistry and TUNEL were used to detect the effects of oncostatin 19 on proliferation and apoptosis of SKOV3 cells and the expression of caspase 3 protein. Results The inhibitory rate of 19 peptide group was 36.47%, the apoptosis of tumor cells was obvious, and the expression of caspase3 protein was significantly increased. The difference was statistically significant compared with the control group (P <0.05). Conclusion Tumor suppressor 19 peptide can significantly inhibit the growth of ovarian cancer cells in nude mice and its mechanism of inhibiting ovarian cancer may be related to its activation of caspase pathway.
