目的利用siRNA靶向沉默p65基因,观察其对三阴乳腺癌细胞系MDA-MB-468、MDA-MB-453体外增殖的影响,从而证实p65对三阴乳腺癌细胞系影响的普遍性。方法以脂质体Lipofectamine TM3000为载体,将针对p65基因的特异性靶点转入人乳腺癌细胞MDA-MB-468、MDA-MB-453细胞;realtime-PCR法检测p65mRNA的表达变化;细胞计数法检测细胞增殖。结果 realtime-PCR检测结果显示,p65siRNA转染组相对于对照组p65mRNA表达显著下降(P<0.05);细胞增殖显示,沉默p65基因后,细胞出现生长抑制现象。结论 siRNA靶向沉默p65基因可抑制人乳腺癌细胞增殖,推测p65对三阴乳腺癌细胞系影响的具有普遍性。
Objective To detect the effect of p65 on the proliferation of human triple-negative breast cancer cell lines by observing the effect of p65 gene silencing on the proliferation of three negative breast cancer cell lines MDA-MB-468 and MDA-MB-453. Methods Liposomes were transfected into human breast cancer cell lines MDA-MB-468 and MDA-MB-453 using lipofectamine TM3000 as a vector. The expression of p65 mRNA was detected by realtime-PCR. Method to detect cell proliferation. Results The results of realtime-PCR showed that the expression of p65 mRNA in p65siRNA transfected group was significantly decreased compared with that in control group (P <0.05). The cell proliferation showed that the cell growth inhibited after p65 gene silencing. Conclusion siRNA targeted silencing of p65 gene inhibits the proliferation of human breast cancer cells. It is presumed that the effect of p65 on the three negative breast cancer cell lines is universal.
