目的:比较复方维甲酸注射液及其拆方制剂青蒿油注射液、维甲酸注射液在体外对人红白血病细胞株K562的增殖、分化和凋亡的影响。方法:用MTT法了解三种注射液对人红白血病细胞株K562增值的抑制作用,用形态学观察、NBT还原实验以及流式细胞术检测表面抗原CD14、CD33观察三种注射液对K562细胞的诱导分化作用;通过流式细胞术观察药物对细胞凋亡和周期的影响。结果:MTT结果显示复方维甲酸注射液对K562细胞增殖有抑制作用,且效果优于其拆方制剂,IC50为11.98±0.06μg/m l;形态学观察、NBT还原实验以及流式检测CD14、CD33均提示复方维甲酸注射液有较好的诱导细胞分化成熟能力;流式检测提示复方维甲酸注射液可将细胞周期阻滞在G2/M期,对K562细胞有诱导早期凋亡的作用,且效果最优。结论:复方维甲酸注射液对人红白血病细胞K562有抑制增殖、诱导分化和凋亡的作用,且效果优于其拆方制剂。
OBJECTIVE: To compare the effects of compound tretinoin injection and its extract preparation of artemisia annua oil injection and retinoic acid injection on the proliferation, differentiation and apoptosis of human erythroleukemia cell line K562 in vitro. Methods: MTT assay was used to investigate the inhibitory effects of three injections on proliferation of human erythroleukemia cell line K562. Morphological observation, NBT reduction assay and flow cytometry were used to detect the surface antigens CD14 and CD33. Induce differentiation; observe the effects of drugs on apoptosis and cycle by flow cytometry. Results: The MTT results showed that the compound tretinoin injection could inhibit the proliferation of K562 cells, and the effect was better than that of the disassembled preparation. The IC50 was 11.98 ± 0.06μg / ml. The morphological observation, NBT reduction assay and flow cytometry analysis of CD14, CD33 Both of them suggest that compound tretinoin injection has good ability of inducing cell differentiation and maturation. Flow cytometry showed that compound tretinoin injection could block cell cycle in G2 / M phase and induce early apoptosis in K562 cells Best effect. CONCLUSION: Compound tretinoin injection can inhibit the proliferation, induce differentiation and apoptosis of human erythroleukemia cell line K562, and its effect is better than its disassembled formulation.
