羟基喜树碱对培养的兔晶状体上皮细胞的影响

目的 探讨羟基喜树碱 (hydroxycamptothecine ,HCPT)对兔晶状体上皮细胞 (rabbitlensepithelialcells ,RLECs)细胞周期的阻断作用和诱导细胞凋亡的作用 ,并研究其对Fas/FasL表达的影响。方法 使用流式细胞仪分析检测HCPT对RLECs细胞周期的阻断作用 ,以光镜和电子显微镜观察细胞形态的变化 ,应用末端脱氧核酸转移酶介导的脱氧三磷酸尿苷缺口末端标记法检测HCPT诱导的细胞凋亡情况 ,应用免疫组化方法检测HCPT对Fas/FasL表达的影响。结果 HCPT可将RLECs阻断于细胞周期的S期并诱导其凋亡 ;凋亡细胞具有典型的形态 ;脱氧尿苷三磷酸标记显示散在阳性细胞 ,且凋亡细胞的数量随HCPT药物浓度的增加而增多。HCPT对RLECs的Fas/FasL表达无明显影响。结论 HCPT不仅可阻断RLECs的分裂 ,而且可诱导细胞发生凋亡 ;Fas/FasL介导途径未参与HCPT诱发RLECs凋亡的过程 Objective To investigate the effect of hydroxycamptothecine (HCPT) on the cell cycle arrest and apoptosis in rabbit lens epithelial cells (RLECs) and to study its effect on the expression of Fas / FasL. Methods Flow cytometry was used to detect the cell cycle arrest effect of HCPT on RLECs. The morphology of RLECs was observed by light microscopy and electron microscopy. HCPT was detected by terminal deoxynucleotidyl transferase mediated dUTP nick end labeling Induced apoptosis and the effect of HCPT on Fas / FasL expression was detected by immunohistochemistry. Results HCPT blocked RLECs in the S phase of the cell cycle and induced apoptosis. Apoptotic cells showed typical morphology. Deoxyuridine triphosphate (TUNEL) showed scattered positive cells, and the number of apoptotic cells increased with the concentration of HCPT And more. HCPT had no significant effect on the expression of Fas / FasL in RLECs. Conclusion HCPT can not only block the division of RLECs, but also induce cell apoptosis; Fas / FasL-mediated pathway is not involved in the process of HCPT-induced apoptosis of RLECs
