目的研究钩端螺旋体Loa22蛋白血清阻断钩体黏附Raw264.7细胞的情况。方法钩体外膜蛋白Loa22、PBS免疫豚鼠后取血清,PBS为对照,使用镀银染色法检测各免疫血清阻止钩体黏附Raw264.7细胞且测定黏附率。结果稀释度为1:200~1:1 600的Loa22蛋白血清均显著抑制钩体黏附,粘附率≤0.01%。而相应稀释度的对照组血清不影响钩体黏附。结论 Loa22蛋白可以阻断钩体黏附细胞,对细胞起到保护作用。
Objective To study the situation that Leptospira Loa22 protein serum block the adhesion of Leptospira to Raw264.7 cells. Methods The outer membrane protein Loa22 of leptospira was isolated from guinea pigs and immunized guinea pigs with serum and PBS as control. The immunofluorescent serum was used to detect the adhesion of Raw264.7 cells and the adherence rate was determined by silver staining. Results Serum Loa22 with a dilution of 1: 200 to 1: 1 600 significantly inhibited leptospiral adhesion with an adhesion rate of ≤0.01%. The corresponding dilution of the control serum does not affect the hook body adhesion. Conclusion Loa22 protein can block the adhesion of leptospira cells, play a protective role on the cells.
