目的:通过实验,建立一种测定穿琥宁的新方法。方法:在pH4.5的酸性溶液中,取适量穿琥宁溶液和伊文思蓝溶液混合,以二次蒸馏水稀释,放置一段时间后在荧光光度计上以λem=λex方式进行同步扫描。结果:伊文思蓝和穿琥宁能够迅速反应生成离子缔合物,导致共振瑞利散射(RRS)光谱的改变,产生比较特征的新的RRS光谱,最大峰值在717nm,并且在25~900mg·L-1范围内,RRS强度和穿琥宁的浓度成正比。结论:我们建立了测定穿琥宁的共振瑞利散射法。该法灵敏度较高,检测限可达到45ng·mL-1,用于检测某些穿琥宁样品中穿琥宁的含量,结果较满意。
Objective: To establish a new method for determining Chuanhuning through experiments. METHODS: In an acidic solution of pH 4.5, appropriate amounts of the potassium dehydroandrographolide solution and the Evans blue solution were mixed and diluted with distilled water. After a period of time, they were simultaneously scanned in the λem=λex mode on a fluorometer. RESULTS: Evans Blue and Chuanhuning could rapidly react to form ionic associates, leading to changes in Resonance Rayleigh Scattering (RRS) spectra, yielding new RRS spectra of comparable characteristics, with peaks at 717 nm and 25 to 900 mg·L- Within the range of 1, RRS intensity is proportional to the concentration of Chuanhuning. Conclusion: We have established a resonance Rayleigh scattering method for the determination of Chuanhuning. The sensitivity of this method is high, and the detection limit can reach 45 ng·mL-1. It is used to detect the content of Chuanhuning in some Chuanhuning samples. The results are satisfactory.
