目的探讨及分析透明细胞肾细胞癌(ccRCC)长链非编码RNA(lncRNA)的表达谱,并进行初步验证。方法利用高通量lncRNA芯片技术分别检测3例ccRCC组织及对应癌旁组织中lncRNA表达谱的差异,对芯片结果进行基因本体论(Gene Ontology)分析,Pathway分析,建立lncRNA与靶基因的基因网络图,并利用实时荧光定量RT-PCR技术对高通量lncRNA芯片检测结果进行验证。结果 ccRCC患者癌组织与癌旁对照样本差异表达的lncRNA共3 862条,其中1 619条上调表达,2 243条下调表达(差异表达倍数≥2.0,P<0.05)。荧光定量RT-PCR结果显示,筛选出的6个lncRNA,在87例ccRCC样本中的表达差异与芯片检测结果一致。结论ccRCC存在明显的lncRNAs差异性表达,这些lncRNAs及某些信号通路可能在肾癌发生和发展中发挥重要作用。
Objective To investigate and analyze the expression profile of long-chain non-coding RNA (lncRNA) in clear cell renal cell carcinoma (ccRCC). Methods The mRNA expression profiles of lncRNA in 3 cases of ccRCC tissues and its corresponding paracancerous tissues were detected by high-throughput lncRNA microarray. Gene Ontology analysis and Pathway analysis were performed on the chip results to establish the gene network of lncRNA and target genes The real-time fluorescence quantitative RT-PCR technique was used to verify the results of high throughput lncRNA microarray assay. Results There were 3 862 lncRNAs differentially expressed in both CCRCC patients and adjacent non-cancerous counterparts, of which 1 619 were up-regulated and 2 243 were down-regulated (difference in expression multiple ≥ 2.0, P <0.05). Fluorescent quantitative RT-PCR results showed that the expression of 6 lncRNAs in 87 samples of ccRCC was consistent with that of the chip. Conclusions There are obvious differences in expression of lncRNAs in ccRCC. These lncRNAs and some signaling pathways may play an important role in the development and progression of renal cell carcinoma.
