[目的]研究乳腺癌组织中金属蛋白酶组织抑制因子 (TIMP_1)的表达状况及其与Ⅳ型胶原 (ColⅣ )的分布形式的关系 ,揭示乳腺癌的浸润转移机制 ,同时为可能应用新治疗方法提供理论依据。[方法]应用SP免疫组化方法检测95例乳腺癌组织和30例乳腺非癌组织中TIMP_1的表达情况和ColⅣ的分布形式。[结果] (1)在95例乳腺癌组织中 ,TIMP_1的阳性表达率为32.6%(31/95) ;ColⅣ分布形式中包裹型占14.7%(14/95)、崩解型占85.3%(81/95)。与对照组比较均有显著差异 (P<0.01)。(2)TIMP_1的表达与组织学分级有关 (Ⅰ级/Ⅲ级 ,P<0.05) ,级别越高 ,阳性率越高。(3)TIMP_1的表达与ColⅣ的分布形式相关(P<0.05) ,TIMP_1阳性者 ,则崩解型ColⅣ出现的机率小。[结论]乳腺癌TIMP_1的表达与肿瘤浸润或转移呈负相关 ,TIMP_1则通过下调MMP对ColⅣ的降解活性而抑制肿瘤细胞的侵袭 ,这为应用MMP抑制剂治疗乳腺癌提供客观依据
[Objective] To investigate the expression of tissue inhibitor of metalloproteinase (TIMP-1) in breast cancer and its relationship with the distribution of type Ⅳ collagen (Col Ⅳ), to reveal the mechanism of invasion and metastasis of breast cancer and to provide new therapeutic methods Theoretical basis. [Methods] The expression of TIMP_1 and the distribution of Col Ⅳ in 95 cases of breast cancer and 30 cases of non-cancerous breast tissues were detected by SP immunohistochemistry. [Results] (1) In 95 cases of breast cancer, the positive expression rate of TIMP_1 was 32.6% (31/95), while that of Col Ⅳ was 14.7% (14/95) and disintegration was 85.3% 81/95). Compared with the control group, there were significant differences (P <0.01). (2) The expression of TIMP_1 was correlated with histological grade (grade Ⅰ / Ⅲ, P <0.05). The higher the grade, the higher the positive rate. (3) The expression of TIMP_1 was correlated with the distribution of Col Ⅳ (P <0.05). The positive rate of ColⅣ was lower in patients with TIMP_1 positive. [Conclusion] The expression of TIMP-1 in breast cancer is negatively correlated with tumor invasion or metastasis. TIMP-1 inhibits the invasion of tumor cells by down-regulating the degradation activity of ColⅣ, which provides an objective basis for the application of MMP inhibitor in the treatment of breast cancer
