目的:研究加味芎归汤对大鼠全脑缺血再灌注损伤的保护作用。方法:采用SD大鼠,以阻断椎动脉血流、双侧颈总动脉阻断再灌注法制备全脑缺血再灌注损伤模型;加味芎归汤(0.38,0.76,1.52 g.kg-1)ig,共9 d,另设模型、假手术和阳性药(通脉颗粒)对照;按NSS法进行神经功能缺损评分,放免法检测血浆中肌酸激酶(CK)与乳酸脱氢酶(LDH)的活性,检测血浆和脑组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,免疫组化法检测脑组织中半胱氨酸天氨酸蛋白酶(caspase-3)的表达,显微镜下进行脑组织病理学检查,称重法测定脑组织含水量。结果:加味芎归汤使脑损伤大鼠脑组织病理改变减轻,脑组织含水量降低,神经功能障碍评分明显下降;脑组织和血浆SOD活性恢复,MDA含量下降,血浆CK和LDH活性降低;脑神经细胞caspase-3表达下降。结论:加味芎归汤对大鼠全脑缺血再灌注损伤有明显保护作用。
Objective: To study the protective effect of Jiawei Xionggui Tang on global cerebral ischemia-reperfusion injury in rats. Methods: SD rats were used to occlude vertebral artery blood flow and occlusion and reperfusion injury of bilateral common carotid artery to establish the model of global cerebral ischemia-reperfusion injury. Modified Xionggui Tang (0.38,0.76,1.52 g.kg-1 ) ig, a total of 9 days, another model, sham operation and positive drug (Tongmai granule) control; NSS neurological deficit score, radioimmunoassay plasma creatine kinase (CK) and lactate dehydrogenase ), The activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) in plasma and brain tissue were detected. The expression of caspase-3 in brain tissue was detected by immunohistochemistry , Under the microscope for histopathological examination, determination of brain water content by weighing method. Results: Modified Xionggui Decoction reduced the pathological changes of brain tissue in rats with brain injury, decreased the water content of brain tissue and decreased the score of neurological dysfunction. The SOD activity of brain tissue and plasma recovered, the content of MDA decreased and the activity of plasma CK and LDH decreased. Brain Neuronal caspase-3 expression decreased. Conclusion: Jiawei Xionggui Tang has a significant protective effect on global cerebral ischemia-reperfusion injury in rats.
