目的:研究细胞间黏附分子-1469K>E基因多态性位点等位基因分布频率及其与2型糖尿病视网膜病变的关系。方法:用PCR检测102例糖尿病视网膜病变患者细胞间黏附分子-1469KK、KE及EE基因型出现的频率,并检测其空腹血糖、三酰甘油、糖化血红蛋白、血浆游离细胞间黏附分子-1和总胆固醇水平,并与120例无糖尿病视网膜病变患者和150例正常对照组比较。结果:2型糖尿病患者空腹血糖、糖化血红蛋白、总胆固醇、三酰甘油、低密度脂蛋白胆固醇、血浆游离细胞间黏附分子-1水平均明显高于正常对照组(P<0.01),高密度脂蛋白胆固醇水平均明显低于正常对照组(P<0.01);糖尿病视网膜病变组总胆固醇、三酰甘油、血浆游离细胞间黏附分子-1水平明显高于无糖尿病视网膜病变组(P<0.01)。KK,KE,EE3种基因型在3组中的分布频率差异有统计学意义(χ2=7.915,P<0.01),糖尿病视网膜病变组等位基因E出现的出现频率明显高于正常对照组和无糖尿病视网膜病变组(P<0.05),2型糖尿病组细胞间黏附分子-1469E等位基因携带者血浆游离细胞间黏附分子-1水平明显高于K携带者(P<0.01)。结论:糖尿病视网膜病变的发生与脂代谢紊乱及细胞间黏附分子-1469K>E基因多态性有关。
OBJECTIVE: To study the allele distribution of intercellular adhesion molecule-1469K> E polymorphism and its relationship with type 2 diabetic retinopathy. Methods: The frequencies of intercellular adhesion molecule-1469KK, KE and EE genotypes in 102 patients with diabetic retinopathy were detected by PCR and their fasting blood glucose, triglyceride, glycosylated hemoglobin, plasma free intercellular adhesion molecule-1 Cholesterol level, and with 120 cases of diabetic retinopathy and 150 normal control group. Results: The levels of fasting blood glucose, glycosylated hemoglobin, total cholesterol, triglyceride, low density lipoprotein cholesterol and plasma intercellular adhesion molecule-1 in type 2 diabetic patients were significantly higher than those in normal controls (P <0.01) (P <0.01). The levels of total cholesterol, triglyceride and plasma intercellular adhesion molecule-1 in diabetic retinopathy group were significantly higher than those in non-diabetic retinopathy group (P <0.01). There were significant differences in the distribution frequency of KK, KE and EE genotypes among the three groups (χ2 = 7.915, P <0.01). The frequency of allele E in diabetic retinopathy group was significantly higher than that in normal control group and no Diabetic retinopathy group (P <0.05), type 2 diabetes mellitus-1469E allele carrier plasma intercellular adhesion molecule-1 levels were significantly higher than K carriers (P <0.01). Conclusion: The incidence of diabetic retinopathy is related to the disturbance of lipid metabolism and the polymorphism of intercellular adhesion molecule -1469K> E.
