目的:分析和比较海南和四川不同生长年限曼地亚红豆杉3种紫杉烷类活性成分的的含量差异。方法:利用甲醇和CHCl3混合液超声浸提后再用甲醇溶解得到相关成分;通过高效液相色谱法对10-DABⅢ、三尖杉宁碱和紫杉醇含量进行测定和比较。结果:1)不同生长年限,海南栽培曼地亚红豆杉3年生枝条中10-DABⅢ和紫杉醇含量均最高,7年生枝条中三尖杉宁碱含量最高;四川栽培曼地亚红豆杉4年生枝条中10-DABⅢ和三尖杉宁碱含量均最高,紫杉醇含量在不同生长年限间不存在显著性差异(P>0.05)。2)相同生长年限,四川栽培曼地亚红豆杉10-DABⅢ含量普遍高于海南栽培曼地亚红豆杉,三尖杉宁碱含量两个产地间有高有低,海南栽培3年生曼地亚红豆杉紫杉醇含量在所有年限间最高。结论:四川栽培曼地亚红豆杉10-DABⅢ含量高于海南栽培曼地亚红豆杉,可以优先作为10-DABⅢ提取的材料来源产地;海南栽培4年生和3年生曼地亚红豆杉分别作为三尖杉宁碱和紫杉醇粗提原料来源更优。
OBJECTIVE: To analyze and compare the contents of three taxanes in Taxus chinensis under different growing seasons in Hainan and Sichuan. Methods: The components were eluted with methanol and CHCl3 and then dissolved in methanol. The contents of 10-DABⅢ, cephalomannine and paclitaxel were determined and compared by HPLC. The results showed that: 1) The content of 10-DABⅢ and paclitaxel in the branches of three-year-old Taxus mairei cultivated in Hainan Province were the highest in different growth years, the content of apixnine was the highest in 7-year-old branches; The contents of 10-DABⅢ and Cephalomannine were the highest, while there was no significant difference in the content of paclitaxel between different growth years (P> 0.05). 2) The same growth period, the cultivation of Taxus mairei 10-DAB Ⅲ content is generally higher than the Hainan cultivation of Taxus cuspidata, Cephalomannine content of the two producing areas have high and low, Hainan cultivation 3-year-old mandela Taxus paclitaxel content in all years, the highest. Conclusion: The content of 10-DABⅢ of Taxus mairei in Sichuan Province is higher than that of Taxus mairea cultivated in Hainan, which can give priority to the origin of 10-DAB Ⅲ extraction materials. The 4-year-old and 3-year-old Taxus mairei cultivars in Hainan Province, Apex cinnabar and paclitaxel crude extract raw materials better.
