目的探讨SDS法、Triton法及胰酶法对肺结核分枝杆菌DNA的检测效果。方法观察3种方法对16例肺结核分枝杆菌DNA提取结果检出的敏感性。结果16份痰液标本,SDS法的阳性为11例(68.08%),Triton法的阳性为13例(81.3%),胰酶法的法阳性为12例(75.0%)。结果显示SDS法的敏感度为2-4,Triton法的敏感度为2-7,胰酶法的敏感度为2-6。Triton法和酶法优于SDS法。但差异无显著性(P>0.05)。结论提示这3种肺结核分枝杆菌DNA制备方法均能检出临床标本中的肺结核分枝杆菌,并具有快速、简便的特点。
Objective To investigate the detection of Mycobacterium tuberculosis DNA by SDS method, Triton method and trypsin method. Methods The sensitivity of three kinds of methods to DNA extraction from 16 cases of Mycobacterium tuberculosis was observed. Results The positive results of SDS method were 11 cases (68.08%) by SDS method, 13 cases (81.3%) by Triton method and 12 cases (75.0%) by trypsin method. The results showed that the sensitivity of SDS method is 2-4, the sensitivity of Triton method is 2-7, and the sensitivity of trypsin method is 2-6. Triton method and enzyme method is superior to SDS method. But the difference was not significant (P> 0.05). Conclusion These results suggest that all three kinds of Mycobacterium tuberculosis DNA preparation methods can detect Mycobacterium tuberculosis in clinical samples and have the characteristics of quickness and simplicity.
