目的:通过动物体内实验验证加味牛蒡子汤(JWNBZT)对类风湿性关节炎的治疗作用。方法:将20只3月龄左右TNF-Tg小鼠按照体重随机分为模型组和加味牛蒡子汤治疗组,另取10只同窝对照体重相似的野生型小鼠(WT)作为空白对照组,治疗组每天以加味牛蒡子汤灌胃,空白对照组和模型组每天以等量纯净水灌胃,治疗12周后测量小鼠踝关节周长和活动度,并取踝关节制作石蜡切片进行HE染色观察炎性浸润和关节结构改变情况。结果:与WT小鼠相比,TNF-Tg小鼠踝关节明显肿胀(P<0.05),踝关节背伸活动度受限;而加味牛蒡子汤治疗12周明显减轻TNF-Tg小鼠踝关节肿胀(P<0.05)和踝关节活动度(P<0.05)。踝关节石蜡切片HE染色显示空白对照组小鼠踝关节结构完整,没有炎性浸润和骨性侵蚀;模型组小鼠踝关节炎性浸润大量产生,骨侵蚀严重,关节结构明显破坏;加味牛蒡子汤组治疗后炎性浸润和骨结构破坏明显减轻。结论:加味牛蒡子汤能够减轻TNF-Tg关节水肿和结构破坏,是炎症性关节炎治疗的潜在替代药物。
OBJECTIVE: To verify the therapeutic effect of flavored Arctium lappa (JWNBZT) on rheumatoid arthritis by in vivo experiments in animals. Methods: 20 3-month-old TNF-Tg mice were randomly divided into model group and modified Arctium lappa decoction group. Ten wild-type mice (WT) with similar litter size were used as blank control group . The treatment group was fed with Fructus Arctii decoction daily. The blank control group and model group were orally administered with the same amount of pure water everyday. After 12 weeks of treatment, the ankle circumference and mobility of the mice were measured. The ankle joint was made into paraffin sections HE staining was used to observe inflammatory infiltration and joint structure changes. Results: Compared with WT mice, the ankle joint of TNF-Tg mouse was significantly swollen (P <0.05) and the mobility of ankle dorsiflexion was limited. However, 12-week treatment of Fructus Arctii decreased TNF-Tg mouse ankle joint Swelling (P <0.05) and ankle joint activity (P <0.05). Ankle joint paraffin sections HE staining showed that the blank control group mice ankle joint structure integrity, no inflammatory infiltration and bone erosion; model group mice ankle infiltration of a large number of generated, severe erosion of the bone, joint structure was significantly damaged; Modified Arctium Soup group inflammatory infiltration and destruction of bone structure significantly reduced. Conclusion: Fructus Arctium decoction can reduce the edema and structure of TNF-Tg joints and is a potential substitute for the treatment of inflammatory arthritis.
