L-精氨酸在过量水杨酸钠致豚鼠内耳损伤中的保护作用

目的观察L -精氨酸在过量水杨酸钠致豚鼠内耳损伤中的作用。方法①将 48只健康且耳廓反射正常的花色豚鼠随机分为四组 :A组 :对照组 (给等量生理盐水 ) 1 2只 ;B组 :水杨酸钠 [45 0mg/(kg·d)组 ;C组 :水杨酸钠 [45 0mg/(kg·d) ]+L -精氨酸 [2 0 0mg/(kg·d) ]组 ;D组 :水杨酸钠 [45 0mg/(kg·d) ]+L -精氨酸 [2 0 0mg/(kg·d) ]组 +亚硝基左旋精氨酸甲酯 [L -NAME 3mg/(kg·d) ]。②测试ABR听反应阈和Ⅰ波潜伏期。③以磷酸二酯酶测定法检测耳蜗组织CaM活性。④用比色测量血浆中的NO含量。结果①各给药组给药后的第 2、4和第 6天ABR阈值均有明显提高 ,对照组ABR阈值没有明显改变。B组给药后阈值抬高最明显 ,于给药后的第 6天达最高。同期各给药组组间比较发现C组用药后第 4、6、8、1 0天ABR阈值明显低于B组和D组 (P <0 .0 5 )。②A组在各时间段所测ABRⅠ波潜伏期无显著波动。B组、D组在给药后的第 4、6、8、1 0天ABRⅠ波潜伏期与同期对照组比较有显著延长 (P <0 .0 5 ) ,这种变化在水杨酸钠给药组尤为明显 (P <0 .0 1 )。③L -Arg治疗组豚鼠耳蜗组织CaM活性明显低于水杨酸钠组 (P <0 .0 1 )及水杨酸钠 +L -Arg +L -NAME组 (P <0 .0 1 )。而血浆NO含量水杨酸钠组明显低于对照组 (P <0 .0 1 )和水杨酸钠 Objective To observe the role of L - arginine in the damage of inner ear of guinea pig induced by excessive amount of sodium salicylate. Methods ①A total of 48 guinea pigs with normal auricle reflexes were randomly divided into four groups: group A: control group (equal to normal saline) 12; group B: sodium salicylate 45Omg / kg Group C: Sodium salicylate [45 0 mg / (kg · d)] + L-arginine [200 mg / (kg · d)]; Group D: Sodium salicylate [45 0 mg / (kg · d)] + L-arginine [200 mg / (kg · d)] group + nitroso L-arginine methyl ester [L -NAME 3 mg / (kg · d)]. ② test ABR listening threshold and Ⅰ wave latency. ③ phosphodiesterase activity assay cochlear CaM activity. ④ with colorimetric measurement of plasma NO content. Results ① The thresholds of ABR were significantly increased on the 2nd, 4th, and 6th day after administration in each treatment group, while the ABR threshold of the control group did not change significantly. After the administration, the threshold elevation in group B was the most obvious, reaching the highest on the 6th day after administration. During the same period, the ABR thresholds of the administration groups were significantly lower than those of the B and D groups on the 4th, 6th, the 10th and the 10th days after the administration of C in the same period (P <0.05). ② There was no significant fluctuation of ABRⅠ wave latency in group A at each time period. In group B and group D, the latency of ABRⅠ wave was significantly prolonged on the 4th, 6th, 10th and 10th day after administration compared with the control group (P <0.05) Group was more obvious (P <0. 01). ③ The CaM activity of cochlear in L-Arg treatment group was significantly lower than that of sodium salicylate group (P <0.01) and sodium salicylate + L -Arg + L -NAME group (P <0.01).
