为了解实验性肝纤维化间质胶原酶(MMP1)基因表达的动态变化,建立了四氯化碳中毒性及人血白蛋白免疫损伤性2种大鼠肝纤维化模型。采用逆转录定量PCR方法,测定肝纤维化不同阶段间质胶原酶基因表达水平。结果表明:正常大鼠肝组织有间质胶原酶的基因表达。在肝纤维化的发生、发展过程中,肝组织间质胶原酶基因表达逐渐增强;到肝硬化阶段MMP1基因表达较肝纤维化阶段下降;在肝硬化的自发逆转过程中MMP1基因表达保持在明显高于正常对照的水平。提示肝纤维化过程中MMP1基因表达与酶活性的变化一致,酶活性的变化是其基因表达改变的结果
To understand the dynamic changes of the gene expression of experimental hepatic fibrosis interstitial collagenase (MMP1), two models of liver fibrosis induced by carbon tetrachloride and human immunoglobulin of immunosuppression were established. Reverse transcription polymerase chain reaction (RT-PCR) was used to determine the gene expression of plasminogen in different stages of hepatic fibrosis. The results showed that the gene expression of interstitial collagenase in normal rat liver tissue. During the development of hepatic fibrosis, the expression of collagenase gene in liver tissue gradually increased; the expression of MMP1 gene in liver cirrhosis stage was lower than that in hepatic fibrosis stage; the expression of MMP1 gene was maintained in the spontaneous reversal of liver cirrhosis Higher than the normal control level. Suggesting that MMP1 gene expression during liver fibrosis consistent with changes in enzyme activity, changes in enzyme activity is the result of changes in their gene expression
