目的:采用近红外(NIR)光谱技术建立一种快速测定白芍药材芍药苷的方法。方法:运用HPLC测定样品中芍药苷的含量,运用偏最小二乘(PLS)法建立含量如NIR光谱之间的多元校正模型,对未知样品进行含量预测。结果:校正集的内部交差验证均方差RMSECV为0.019 7,校正模型中真实值与预测值之间的相关系数(R2)为0.969 6,外部验证均方差(RMSEP)为0.046 1。结论:近红外漫反射光谱法快速、简便、无损,可用于白芍药材中芍药苷含量的快速检测,对其他药材中指标性成分的测定也有一定的参考价值。
Objective: To establish a rapid method for the determination of paeoniflorin in Paeonia lactiflora by near infrared spectroscopy (NIR). Methods: The contents of paeoniflorin in the samples were determined by HPLC, and multivariate calibration models such as NIR spectra were established by partial least squares (PLS) method to predict the content of the unknown samples. Results: The mean square error RMSECV of cross validation in the calibration set was 0.019 7. The correlation coefficient (R2) between the true value and the predicted value in the calibration model was 0.969 6, and the RMSEP was 0.046 1. Conclusion: NIR diffuse reflectance spectrometry is rapid, simple and non-destructive. It can be used to detect the content of paeoniflorin in Radix Paeoniae Alba, and also has certain reference value for the determination of the index components in other medicinal materials.
