目的 :探讨以国产免疫磁珠进行细胞分离的方法 ,并评价直接包被法的特异性和敏感性及其在脐血和骨髓 CD+ 34 细胞分离中的应用。方法 :直接包被法、SPA法、生物素 -亲和素法。结果 :直接包被法、SPA法和生物素-亲合素法均可有效分离细胞 ,但以直接包被法最具优势 ,并证明其有较高特异性和敏感性。应用直接包被法分离脐血及骨髓中的 CD+ 34 细胞率为 5 1%~ 82 % ,台盼蓝拒染率为 82 %~ 88%。结论 :国产免疫磁珠有较好的分离效果 ,对细胞活性影响较小
OBJECTIVE: To investigate the method of cell separation with domestic immunomagnetic beads and to evaluate the specificity and sensitivity of direct coating method and its application in the isolation of cord blood and bone marrow CD + 34 cells. Methods: direct coating method, SPA method, biotin-avidin method. Results: Direct coating method, SPA method and biotin - avidin method all could effectively separate cells, but the direct coating method had the most advantage and proved to have higher specificity and sensitivity. The rate of CD + 34 cells in cord blood and bone marrow was 51.1% ~ 82% and the trypan blue exclusion rate was 82% ~ 88%. Conclusion: The domestic immunomagnetic beads have better separation effect, less impact on cell activity
