目的以肠溶性的醋酸羟丙基甲基纤维素琥珀酸酯(HPMCAS)作为包覆材料,包覆于负载了抗癌药物与5-氟脲嘧啶(5-Fu)的SBA-15介孔分子筛表面,制备pH敏感的药物控释系统HPMCAS/SBA-15。方法考察HPMCAS/SBA-15系统在模拟胃液(pH=1.2)和模拟肠液(pH=7.5)中的释放行为。结果在模拟胃液中,HPMCAS能够明显地延缓5-Fu的释放速度,药物释放4 h后,其释放率仅为14.3%,而在模拟肠液中,HPMCAS能够迅速溶于肠液,对5-Fu释放速度的影响甚微,药物释放4 h后,释放率可达到83.7%。结论 HPMCA/SBA-15是一种良好的pH敏感型药物缓解材料。
Objective To prepare HPMCAS coated with SBA-15 mesoporous molecular sieves loaded with anticancer drug and 5-fluorouracil (5-Fu) Surface preparation of pH-sensitive drug delivery system HPMCAS / SBA-15. Methods The release behavior of HPMCAS / SBA-15 system in simulated gastric fluid (pH = 1.2) and simulated intestinal fluid (pH = 7.5) was investigated. Results In simulated gastric fluid, HPMCAS could delay the release rate of 5-Fu significantly. After 4 h, the release rate of HPMCAS was only 14.3%. In simulated intestinal fluid, HPMCAS could dissolve rapidly in intestinal fluid and release 5-Fu The effect of speed was minimal, and the release rate reached 83.7% after 4 h of drug release. Conclusion HPMCA / SBA-15 is a good pH-sensitive drug remediation material.
