目的:探讨经47℃20分钟短时超高温处理后,胃癌细胞和正常胃组织细胞在体外培养的效应,以及细胞对某些抗癌药物的敏感性。方法:将新鲜胃癌细胞分别经43℃120分钟或47℃20分钟热处理后,进行体外抗癌药敏培养,并按MTT法检测细胞生长抑制率,与无热处理的胃癌细胞和正常胃组织培养结果相比较,以F检验判断其显著性。结果:43℃120分钟热处理可以明显抑制胃癌细胞在体外的生长,可以增加胃癌细胞对某些抗癌药物的敏感性;两种处理方式对胃癌细胞的影响结果相似,而对正常胃组织影响不明显。结论:47℃20分钟的短时超高温处理,可以达到43℃120分钟的常规热处理的目的。胃癌细胞经短时超高温处理后,可选用ADM、MMC、5-FU、VP-16等抗癌药物。47℃20分钟短时超高温处理不影响正常胃组织细胞在体外的生长
OBJECTIVE: To investigate the effects of in vitro culture of gastric cancer cells and normal gastric tissue cells after a short hyperthermic treatment at 47°C for 20 minutes, and the sensitivity of the cells to certain anticancer drugs. METHODS: Fresh gastric cancer cells were heat-treated at 43°C for 120 minutes or 47°C for 20 minutes and then treated with anti-cancer susceptibility culture in vitro. The cell growth inhibition rate was measured by MTT assay, and compared with that of non-heat-treated gastric cancer cells and normal gastric tissues. In comparison, the F test was used to judge its significance. RESULTS: Heat treatment at 43°C for 120 minutes could significantly inhibit the growth of gastric cancer cells in vitro, and increase the sensitivity of gastric cancer cells to certain anticancer drugs. The effects of the two treatments on gastric cancer cells were similar, but the effects on normal gastric tissues were not affected. obvious. Conclusion: Short-term ultra-high temperature treatment at 47°C for 20 minutes can achieve the purpose of conventional heat treatment at 43°C for 120 minutes. After short-term hyperthermia treatment of gastric cancer cells, ADM, MMC, 5-FU, VP-16 and other anti-cancer drugs can be used.
