目的探讨核因子-κB-p65(Nuclear factor-κB-p65,NF-κB-p65)与基质金属蛋白酶-1(Matrix metalloproteases-1,MMP-1)在非小细胞肺癌(Non-small cell lung cancer,NSCLC)组织中的表达及其与肿瘤浸润、转移、凋亡的相关性。方法收集50例手术切除的NSCLC组织及15例癌旁正常组织标本,RT-PCR法检测NSCLC及癌旁正常组织中NF-κB-p65 mRNA的表达,免疫组化法检测MMP-1蛋白的表达,TUNEL法检测细胞凋亡指数。结果 NSCLC组织中NF-κB-p65 mRNA的表达量显著高于癌旁正常组织(P<0.05);在腺癌组织中的表达量显著高于鳞癌组织(P<0.05);NSCLC组织中MMP-1蛋白阳性率显著高于癌旁正常组织(P<0.01);NF-κB-p65 mRNA和MMP-1蛋白的表达与TNM分期、分化程度、淋巴结转移、病理类型有关(P<0.05或<0.01),与年龄、性别无关(P>0.05);直线相关分析发现,NSCLC组织中NF-κB-p65 mRNA与MMP-1蛋白的表达相关;NSCLC组织中的细胞凋亡指数显著低于癌旁正常组织,且与NF-κB-p65 mRNA的表达量呈负相关(r=-0.547,P<0.001)。结论 NSCLC组织中NF-κB-p65 mRNA与MMP-1蛋白的表达具有相关性,NF-κB-p65 mRNA的表达与细胞凋亡指数呈负相关,NF-κB-p65通过一定的途径调控MMP-1蛋白的表达,NF-κB与MMP-1共同参与了肿瘤发生、发展的过程,NF-κB-p65发挥抗凋亡作用。
Objective To investigate the role of nuclear factor-κB-p65 and matrix metalloproteinase-1 (MMP-1) in non-small cell lung cancer , NSCLC) and its correlation with tumor invasion, metastasis and apoptosis. Methods Fifty specimens of NSCLC and 15 adjacent normal tissues were collected. The expression of NF-κB-p65 mRNA was detected by RT-PCR and the expression of MMP-1 was detected by immunohistochemistry TUNEL method was used to detect the apoptosis index. Results The expression of NF-κB-p65 mRNA in NSCLC tissues was significantly higher than that in adjacent normal tissues (P <0.05), and was significantly higher in adenocarcinoma tissues than in squamous cell carcinoma (P <0.05) (P <0.01). The expression of NF-κB-p65 mRNA and MMP-1 protein were correlated with TNM stage, differentiation, lymph node metastasis and pathological type (P <0.05 or < 0.01), but had no correlation with age and gender (P> 0.05). The linear correlation analysis showed that the expression of NF-κB-p65 mRNA was correlated with the expression of MMP-1 protein in NSCLC tissues; the apoptosis index in NSCLC tissues was significantly lower Normal tissue, and negatively correlated with the expression of NF-κB-p65 mRNA (r = -0.547, P <0.001). Conclusion The expression of NF-κB-p65 mRNA and MMP-1 protein are correlated in NSCLC tissues. The expression of NF-κB-p65 mRNA is negatively correlated with the apoptosis index. NF-κB-p65 regulates the expression of MMP- 1 protein expression, NF-κB and MMP-1 are involved in the process of tumorigenesis, development, NF-κB-p65 play an anti-apoptotic effect.
