目的建立高效液相色谱法测定西伯利亚接骨木中槲皮素、芦丁含量。方法采用HPLC法,色谱柱:Agilent C18(4.6mm×25 0mm,5μm),流动相:甲醇-0.1%磷酸水溶液(49∶51),柱温25℃,流速1.0mL/min,检测波长254nm。结果芦丁在0.2~1mg/mL浓度范围内线性关系良好(r=0.999 9),平均回收率为100.38%,RSD=2.04%(n=9);槲皮素在0.8~8μg/mL浓度范围内线性关系良好(r=0.999 7),平均回收率为99.23%,RSD=0.96%(n=9)。槲皮素含量为0.097 1~0.198 5mg/g,芦丁含量为7.785~13.300mg/g。结论此方法简便、准确、可靠,可作为西伯利亚接骨木药材芦丁、槲皮素含量的测定方法,为完善西伯利亚接骨木的质量标准提供理论依据。
Objective To establish a method for the determination of quercetin and rutin in Siberian sambucus by high performance liquid chromatography. Methods The mobile phase was methanol-0.1% phosphoric acid (49:51). The column temperature was 25 ℃, the flow rate was 1.0 mL / min and the detection wavelength was 254 nm. The column was Agilent C18 (4.6 mm × 250 mm, 5 μm). Results Rutin had a good linear relationship (r = 0.999 9) with the average recovery of 100.38% and RSD 2.04% (n = 9) in the range of 0.2 ~ 1 mg / mL. The linear relationship was good (r = 0.999 7) with an average recovery of 99.23% and RSD = 0.96% (n = 9). Quercetin content of 0.097 1 ~ 0.198 5mg / g, rutin content of 7.785 ~ 13.300mg / g. Conclusion This method is simple, accurate and reliable, which can be used as a method for determination of rutin and quercetin in Siberian elderberry. It provides a theoretical basis for improving the quality standard of Siberian elk.
