目的 建立舌癌远隔脏器转移的细胞系 ,研究其相关生物学特性。方法 采用瘤细胞裸鼠尾静脉注射法 ,细胞培养法获取舌癌远隔脏器转移细胞系 ,用相差显微镜、HE染色、电镜观察其形态及细胞内部结构 ,染色法显示染色体形态 ,裸鼠背部皮下注射瘤细胞法检验致瘤性并证明其性质。用 2 4孔培养板行生长曲线测定 ,并计算其细胞群体倍增时间 ,用流式细胞仪测定细胞周期来检测其细胞增殖速度。结果 从 3 5只受试裸鼠中获得一只截瘫的裸鼠 ,取其肺、肝、脑和脊髓组织分别行原代培养 ,仅脊髓组织培养获得了稳定传代的细胞系 ,传代 5 0次以上 ,生长稳定 ,Ts倍增时间为 2 6.83h ,S期细胞占 2 5 .5 % ,染色体众数为 60 ;其母细胞Tb倍增时间为 2 8.81h ,S期细胞占 2 1.2 % ,染色体众数为 5 0 ,保持人类染色体形态 ,细胞形态与其母细胞相似 ,裸鼠皮下所致肿瘤组织切片显示为典型的鳞状上皮细胞癌 ,将该细胞命名为Ts。结论 Ts细胞是Tb细胞在裸鼠体内的脊髓转移细胞系 ,生物学特性与其母细胞存在差异
Objective To establish a cell line that metastasizes to distant organs of tongue cancer and to study the related biological characteristics. Methods The tumor cells were transplanted into the distant organs of tongue cancer by tail vein injection and cell culture. The morphology and cell internal structure were observed by phase contrast microscopy, HE staining and electron microscopy. Subcutaneous injection of tumor cells test for tumorigenicity and prove its nature. The growth curve of 24-well plates was used to measure the cell population doubling time, and the cell cycle was measured by flow cytometry to detect the cell proliferation rate. Results A paraplegic nude mouse was obtained from 35 experimental nude mice. The lung, liver, brain and spinal cord tissues were primary cultured and only the spinal cord tissue culture was performed to obtain a stable passage cell line, which was passaged 50 times , The growth was stable, the Ts doubling time was 2 6.83h, the S phase cells accounted for 25.5%, the chromosome number was 60; the Tb doubling time of mother cells was 8.81h, the S phase cells accounted for 2 1.2% The number of 5 0, to maintain human chromosome morphology, cell morphology and its mother cells similar to the nude mice subcutaneous tumor tissue sections showed typical squamous cell carcinoma, the cell named Ts. Conclusion Ts cells are the cell line of spinal cord metastasis of Tb cells in nude mice, and their biological characteristics are different from that of their parental cells
