目的研究鼠受精素β胞外区(Fβextracellular domain,F_(β.ECD))真核表达重组质粒pSG.SS.C3d3.YL.F_(β.ECD)的DNA疫苗免疫避孕效应。方法应用pSG.SS.C3d3.YL.F_(β.ECD)(C3d3-YL-Fβ组)肌注雌鼠,观察其免疫避孕效应,ELISA法检测雌鼠血清抗Fβ蛋白IgG抗体最大稀释度、IL-2、INF-γ、IL-4、IL-10水平和阴道分泌物抗Fβ蛋白IgA抗体最大稀释度。对照组为PBS组、空载体pSG.SS.YL组(空载体组)和pSG.SS.YL.Fβ.ECD(YL-Fβ组)。结果C3d3-YL-Fβ组雌鼠生育能力显著降低,雌鼠妊娠率显著降低,一胎产仔数量显著降低,免疫避孕效果显著好于YL-Fβ组。pSG.SS.C3d3.YL.F_(β.ECD)DNA疫苗可激发雌鼠产生高水平血清抗Fβ蛋白IgG抗体,阴道分泌物中出现抗Fβ蛋白IgA抗体,血清IL-2、INF-γ浓度较对照组显著降低,血清IL-4、IL-10浓度显著增加。结论真核表达重组质粒pSG.SS.C3d3.YL.Fβ.ECD可有效诱导雌鼠免疫性避孕效应,效果优于pSG.SS.YL.F_(β.ECD)。
Objective To study the effect of DNA vaccine immunocontraceptive on eosinophil eukaryotic expression recombinant plasmid pSG.SS.C3d3.YL.F_ (β.ECD). Methods Female mice were intramuscularly injected with pSG.SS.C3d3.YL.F_ (β.ECD) (C3d3-YL-Fβ) to observe the effect of contraceptive immunization. The maximum dilutions of anti-Fβ IgG in serum were detected by ELISA. IL-2, INF-γ, IL-4, IL-10 levels and vaginal secretions anti-Fβ protein IgA antibody maximum dilution. The control group was PBS group, empty vector pSG.SS.YL group (empty vector group) and pSG.SS.YL.Fβ.ECD (YL-Fβ group). Results The female fertility of C3d3-YL-Fβ group was significantly decreased, the pregnancy rate of female mice was significantly decreased, the number of litter size was significantly decreased, and the effect of immunocontraception was significantly better than that of YL-Fβ group. The pSG.SS.C3d3.YL.F_ (β.ECD) DNA vaccine induced high levels of serum anti-Fβ IgG antibodies in female mice, anti-Fβ IgA antibody in vaginal secretions, serum IL-2 and INF-γ concentrations Significantly lower than the control group, serum IL-4, IL-10 concentration was significantly increased. Conclusion The eukaryotic expression plasmid pSG.SS.C3d3.YL.Fβ.ECD can effectively induce contraceptive effect of female mice, which is better than pSG.SY.F (β.ECD).
