目的:研究阳和汤对阳虚证Lewis肺癌小鼠抑瘤率、NK细胞活性、Bax、Bcl-2、VEGF水平的影响。方法:将40只小鼠随机分为阳虚Lewis组、阳和汤临床等效剂量组、阳和汤2倍临床等效剂量组、顺铂组,每组10只。采用氢化可的松注射液肌肉注射制作阳虚证小鼠模型,然后将Lewis肺癌细胞株接种于小鼠右侧腋部皮下,造模1周后开始给药。末次给药24 h后处死小鼠,取瘤称重计算抑瘤率,采用乳酸脱氢酶释放实验检测小鼠脾脏NK细胞活性,取瘤组织分别测定Bcl-2、Bax、VEGF的表达。结果:阳和汤对阳虚证Lewis肺癌小鼠肿瘤生长具有抑制作用,能恢复阳虚证Lewis肺癌小鼠脾脏NK细胞活性,有效降低阳虚证Lewis肺癌小鼠Blc-2表达,提高Bax表达,并且能降低VEGF表达。结论:阳和汤对于阳虚证肺癌的生长具有明确抑制作用,其作用强度与剂量有关,其抗肿瘤的可能机制是增加NK杀伤细胞活性、调节Bax、Bcl-2表达水平和抑制肿瘤血管生成。
Objective: To study the effects of Yanghe decoction on the tumor inhibition rate, NK cell activity, Bax, Bcl-2 and VEGF in Lewis Lung Cancer mice with Yang Deficiency Syndrome. Methods: Forty mice were randomly divided into two groups: Yang-Lewis group, Yanghe Decoction equivalent dose group, Yanghe Decoction 2-fold clinical equivalent dose group and Cisplatin group, 10 rats in each group. Hydrocortisone injection intramuscular injection of Yang deficiency syndrome model mice, and then Lewis lung cancer cell lines inoculated on the right mouse axillary subcutaneous, 1 week after the model began to be administered. Twenty-four hours after the last administration, the mice were sacrificed, the tumor-bearing rate was calculated by tumor weight, and the activity of NK cells in spleen of mice was measured by lactate dehydrogenase release assay. The expression of Bcl-2, Bax and VEGF were measured respectively. Results: Yanghe Decoction could inhibit the growth of Lewis lung carcinoma in mice with Yang Deficiency Syndrome, restore the activity of NK cells in Lewis lung cancer mice with Yang deficiency syndrome, and reduce the expression of Bcl-2 in Lewis lung cancer mice with Yang deficiency syndrome , And can reduce VEGF expression. Conclusion: Yanghe Decoction can inhibit the growth of lung cancer of Yang Deficiency Syndrome and its intensity is dose-dependent. The possible mechanism of anti-tumor is to increase the activity of NK killer cells, regulate the expression of Bax and Bcl-2, and inhibit tumor angiogenesis .
