目的 探讨非小细胞肺癌 (NSCLC)Fas蛋白表达与DNA的关系。方法 对 33份手术切除的NSCLC标本和 17份正常肺组织使用流式细胞术检测Fas蛋白表达和DNA含量 ,并与细胞增殖活性、增殖指数 (PI)、S期分数 (SPF)以及临床病理特征之间进行比较分析。结果 NSCLC的异倍体率为82 % ,DNA指数 (DI)及PI、SPF分别为 1 36± 0 2 3,0 2 9± 0 0 9,0 144± 0 0 78,与正常肺组织相比差异有极显著性 (P =0 0 0 0 )。在NSCLC中Fas的表达低于正常肺组织 (P =0 0 0 0 ) ,并显示与PI、SPF呈负相关 (P分别为 0 0 2 4、0 0 0 4)。Fas表达及细胞增殖活性与NSCLC的临床病理特征如年龄、肿瘤大小、淋巴结转移、病理类型之间未见明显相关。结论 NSCLC的Fas表达降低和细胞增殖活性增高具有普遍意义 ;Fas表达的下降影响了NSCLC细胞的增殖活性 ,这种增殖与凋亡的调控异常在NSCLC发病中可能起重要作用。
Objective To investigate the relationship between Fas protein expression and DNA in non-small cell lung cancer (NSCLC). Methods The expression of Fas protein and DNA in 33 NSCLC specimens and 17 normal lung tissues were detected by flow cytometry. The results were compared with the cell proliferation activity, proliferation index (PI), S phase fraction (SPF) and clinicopathological features Between the comparative analysis. Results The aneuploidy rate of NSCLC was 82%. The DNA index (DI), PI and SPF were 136 ± 0 2 3, 0 9 ± 0 0 9, 0 144 ± 0 0 78, respectively. Compared with normal lung tissue The difference was significant (P = 0 0 0 0). The expression of Fas in NSCLC was lower than that in normal lung tissue (P = 0 0 0 0), and was negatively correlated with PI and SPF (P 0 0 2 4 and 0 0 0 4, respectively). There was no significant correlation between the expression of Fas and cell proliferation and the clinicopathological features of NSCLC such as age, tumor size, lymph node metastasis and pathological type. Conclusion The decrease of Fas expression and the increase of cell proliferative activity in NSCLC are of general significance. The decrease of Fas expression affects the proliferative activity of NSCLC cells. The abnormal regulation of proliferation and apoptosis may play an important role in the pathogenesis of NSCLC.
