目的探讨左归丸联合温和灸治疗骨质疏松症的可能作用机制。方法 40只雌性SD大鼠随机分为空白组、模型组、左归丸组、左归丸+温和灸组,每组10只。模型组、左归丸组、左归丸+温和灸组采用手术完整摘除双侧卵巢法制造骨质疏松症模型。造模3个月后左归丸组予左归丸10 ml/(kg·d)灌胃,左归丸+温和灸组予左归丸10 ml/(kg·d)灌胃+长强穴艾灸治疗,空白组和模型组予生理盐水10 ml/(kg·d)灌胃。各组均干预6周。干预前后测定大鼠股骨近端及第2腰椎骨密度,干预后通过PCR法检测大鼠骨组织骨保护素(OPG)mRNA和核因子κB受体活化因子配体(RANKL)mRNA表达水平。结果与空白组比较,模型组股骨、腰椎骨密度,骨组织OPG mRNA表达及OPG/RANKL降低,RANKL mRNA表达上升(P<0.05或P<0.01);与模型组比较,左归丸组、左归丸+温和灸组大鼠股骨、腰椎骨密度,骨组织OPG mRNA表达及OPG/RANKL均升高,RANKL mRNA表达降低(P<0.05或P<0.01),且左归丸+温和灸组大鼠股骨、腰椎骨密度,骨组织OPG mRNA表达及OPG/RANKL均较左归丸组升高,RANKL mRNA表达较左归丸组降低(P<0.05)。结论左归丸联合温和灸能明显升高骨质疏松症大鼠的骨密度,上调OPG mRNA表达、下调RANKL mRNA表达可能是其作用机制之一。
Objective To explore the possible mechanism of Zuogui Pill combined with mild moxibustion in the treatment of osteoporosis. Methods 40 female SD rats were randomly divided into blank group, model group, Zuoguiwan group, Zuoguiwan + mild moxibustion group, with 10 rats in each group. Model group, Zuoguiwan group and Zuoguiwan group + mild moxibustion group were treated with complete ovarian removal by ovariectomy to make osteoporosis model. Zuoguiwan group was given Zuogui Pill 10 ml / (kg · d) orally 3 months after modeling. Zuo Gui Pill + Moxibustion group was given Zuogui Pill 10 ml / (kg · d) Moxibustion treatment, blank group and model group were given normal saline 10 ml / (kg · d) gavage. Each group were intervention for 6 weeks. The BMD of the proximal femur and the second lumbar vertebrae were measured before and after intervention. The expression of osteoprotegerin (OPG) mRNA and RANKL mRNA was detected by PCR after intervention. Results Compared with the blank group, the femoral and lumbar BMD, the expression of OPG mRNA and OPG / RANKL in the model group decreased and the expression of RANKL mRNA increased (P <0.05 or P <0.01). Compared with the model group, The bone mineral density of femur and lumbar, OPG mRNA expression, OPG / RANKL and the expression of RANKL mRNA of the rats in Guizhi + Moxibustion group were significantly decreased (P <0.05 or P <0.01), and Zuogui Pill + mild moxibustion group Compared with Zuoguiwan group, the expression of OPG mRNA and OPG mRNA in the femur and lumbar vertebrae increased, and the expression of RANKL mRNA decreased compared with those in Zuoguiwan group (P <0.05). Conclusion Zuogui Pill combined with mild moxibustion can significantly increase bone mineral density in osteoporosis rats, and increase the expression of OPG mRNA and down regulate the expression of RANKL mRNA may be one of its mechanisms.
