目的:在建立大鼠牙周炎及大鼠怀孕模型基础上,研究牙周炎与生长受限胎鼠之间的相关关系。方法:11只雌性SD大鼠双侧下颌第一磨牙丝线结扎制作牙周炎动物模型,7只雌性SD大鼠作对照组,2周后将雌鼠与雄鼠合笼建立大鼠怀孕模型。分娩前处死孕鼠,收集孕鼠心脏血、胎鼠,测量胎鼠体重、顶臀长度,ELISA方法检测孕鼠血清中肿瘤坏死因子-α(TNF-α)的浓度。结果:牙周炎组中胎鼠生长受限的发生率为8.3%,显著高于对照组胎鼠生长受限的发生率。牙周炎组生长受限胎鼠的孕鼠TNF-α血清浓度显著高于对照组孕鼠血清浓度(P﹤0.05)。结论:牙周炎是胎鼠生长受限发生的危险因素,可能与牙周局部感染引起血液中TNF-a等炎性细胞因子的改变有关。
Objective: To establish a rat model of periodontitis and pregnancy based on the study of periodontitis and growth-restricted fetal relationship between fetuses. Methods: Animal models of periodontitis were established by ligating the bilateral first mandibular first molar of 11 female SD rats. Seven female SD rats were used as the control group. After 2 weeks, the pregnant rats and male rats were caged to establish the pregnant model. The pregnant rats were sacrificed before delivery, and the heart blood and fetus of pregnant rats were collected. The body weight and the length of the top of the hip were measured. The serum concentrations of tumor necrosis factor-α (TNF-α) in pregnant rats were measured by ELISA. Results: The incidence of fetal growth restriction was 8.3% in the periodontitis group, which was significantly higher than that in the control group. The serum concentration of TNF-α in pregnant rats with periodontitis group was significantly higher than that in control group (P <0.05). Conclusion: Periodontitis is a risk factor for fetal growth restriction, which may be related to the changes of inflammatory cytokines such as TNF-a in blood caused by periodontal infection.
