研究骨形成蛋白-2(Bone Morphogenetic Protein 2,BMP-2)对在不同血清浓度条件下OPG-/-小鼠骨髓间充质干细胞(Bone Marrow Derived Stroma Cells,BMSCs)的增殖和分化作用。取OPG-/-小鼠股骨,分离骨髓基质细胞进行体外培养。在各自培养条件下分为实验组(BMP-2组)和空白对照组(CON组),倒置相差显微镜下观察细胞形态变化,MTT法测定细胞增殖率,PNPP法测定碱性磷酸酶(AlkalinePhosphatase,ALP)的表达。BMP-2组和CON组在含有10%、15%和20%FBS的培养条件下,第3、5和7d的增殖较无FBS和5%FBS条件下明显(P<0.05),而15%和20%FBS条件下增殖又较10%FBS显著(P<0.05);BMP-2组对ALP表达的影响与CON存在较大差异,BMP-2组在所有不同血清浓度条件下的第3、5和7 dALP活性均高于CON组(P<0.05)。BMP-2组对OPG-/-小鼠BMSCs无增殖作用,较高血清浓度是其增殖的主要条件,但在ALP的分化活性方面作用显著。
To investigate the effects of Bone Morphogenetic Protein 2 (BMP-2) on the proliferation and differentiation of OPG - / - mouse BMSCs under different serum concentrations of Bone Marrow Derived Stromal Cells (BMSCs). OPG - / - mouse femur was taken and bone marrow stromal cells were isolated and cultured in vitro. The cells were divided into experimental group (BMP-2 group) and blank control group (CON group) under different culture conditions. Cell morphology was observed under inverted phase contrast microscope. Cell proliferation rate was measured by MTT assay. Alkaline phosphatase ALP) expression. The proliferation of BMP-2 and CON groups at 3, 5, and 7d were significantly (P <0.05) higher than those without FBS and 5% FBS under the culture conditions of 10%, 15% and 20% (P <0.05). The effect of BMP-2 on the expression of ALP was significantly different from that of CON. The expression of ALP in BMP-2 group was significantly higher than that in 10% FBS (P < 5 and 7 dALP activity were higher than CON group (P <0.05). BMP-2 group had no effect on proliferation of OPG - / - mice BMSCs. Higher serum concentration was the main condition for its proliferation, but it had a significant effect on the differentiation activity of ALP.
