目的利用基因表达谱芯片技术筛选肝细胞癌(HCC)中表达上调最为明显的基因。方法利用Affymetrix U133 Plus2.0表达芯片检测93例HCC及配对癌旁肝组织的全基因表达谱。HCC与配对癌旁肝组织中基因表达的比较采用配对t检验,筛选表达上调最为明显的10个基因。各基因表达与临床病理参数的关系采用χ~2检验。结果与癌旁肝组织相比,HCC中表达上调倍数最多的10个基因分别为:CDKN3、CCL20、PTTG1、MELK、PRMT1、TOP2A、TPX2、SMYD3、TMEM106C和UCK2,上调25.2~44.6倍(均P≤2.0×10~(-22))。CCL20、PRMT1和UCK2基因表达与血清AFP有关,CCL20、PTTG1、PRMT1、TOP2A和UCK2基因表达与组织学Edmondson-Steiner分级有关,CCL20基因表达与TNM分期有关(均P<0.05)。结论筛选出HCC中表达上调最为明显的10个基因,将为HCC分子靶向治疗研究提供潜在靶点。
Objective To screen the most up-regulated genes in hepatocellular carcinoma (HCC) by gene expression profiling. Methods Affymetrix U133 Plus2.0 expression vector was used to detect the total gene expression profiles of 93 HCC and matched paracancer liver tissues. Comparison of gene expression in HCC and paired para-liver tissue Using paired t-test, 10 genes that were most significantly up-regulated were screened. The relationship between the gene expression and clinical pathological parameters using χ ~ 2 test. Results Compared with the adjacent noncancerous liver tissues, the up-regulated genes in HCC were up-regulated by 25.2-44.6 folds (P <0.05) with CDKN3, CCL20, PTTG1, MELK, PRMT1, TOP2A, TPX2, SMYD3, TMEM106C and UCK2 ≦ 2.0 × 10 ~ (-22)). The expressions of CCL20, PRMT1 and UCK2 were correlated with serum AFP. The expression of CCL20, PTTG1, PRMT1, TOP2A and UCK2 were related to the histological Edmondson-Steiner classification. The CCL20 gene expression was correlated with TNM stage (all P <0.05). Conclusion The 10 genes with the most significant up-regulation in HCC were screened out, which will provide a potential target for HCC molecular targeted therapy research.
